PF TEK AS PRESENTED BY RR IN THE "LET'S GROW MUSHROOMS" VIDEO SERIES GROW #4 for Mycologist217
Objectives: 1. Demonstrate intermediate+ practical knowledge of the PF TEK and all procedures therein.
2. Demonstrate intermediate+ practical knowledge in Liquid Culture as it applies to the PF TEK. 3. Maintain high rate of contaminant free cultivation. 4. Provide successful and accurate example and instruction to beginning mycologists as they apply to the PF TEK. 5. Gain insight to starting for advance grows.
Required Materials 1. 8oz wide mouth glass canning jars w/ lid (27 36 Jars this grow) 2. All natural brown rice flour (1 cup per 5 jars) 3. Fine Grain Vermiculite (2cups per 5 jars + filter layers) 4. Water (1cup per 5 jars+more) 5. Drill with 3/32" drill bit 6. Micropore Medical Tape 7. Aluminum foil (enough for one foil jar lid per jar) 8. Pressure cooker or stockpot 9. Cotton balls (optional, component of homemade alcohol lamp) 10. Paper towels 11. Isopropyl alcohol (91% and 70%) 12. Homemade alcohol flame lamp 13. Disinfectant cleaner with bleach 14. Premade viable mushroom spore solution in syringe (2 unnamed species 1 premade psilocybe cubensis PENIS ENVY, and 3 premade MS solution of unnamed cube at ~1ml per jar) 15. Homemade Still Air Box 16. Homemade SGFC (shotgun fruiting chamber) 17. Perlite (enough for ~4" deep in SGFC) 18. Latex gloves 19. Surgical face mask.
20. Preconstructed LC (liquid culture) jar lids with jars
21. Honey 22. Space heater (optional)
Procedure: this is my 4th attempt at RR's PF tek, and to date I have not had any jar lost to contamination following this TEK. Click HERE for the lLet's Grow Mushrooms PF TEK video tutorial presented by RR.
Day -1: Pre-inoculation Today the last several components of the materials list were assembled, and I began the PF tek, by constructing 24 PF TEK jar lids, filling nine with substrate, and steam sterilizing those same nine jars in my pressure cooker. I constructed my alcohol flame lamp using a small garlic jar, a cotton ball, and 91% isopropyl alcohol. I have also prepared a small room in my house for inoculations tomorrow by thoroughly dusting the area and sanitizing with disinfectant cleaner.
Due to cooling times and limited space in myressure cooker, I am resigned to prepare nine jars for inoculation each day until 27 36 inoculated cultures are produced. The first nine to receive spore tomorrow will be a new strain for me whilst the latter 18 jars will be a strain I have grown using the PF TEK before. I will be providing pictures througnout the entirety of this experiment starting tomorrow with my inoculation room set up. Day 0. The first of three inoculation rounds was executed this morning. I observed that water droplets made their way onto the top of at least 2/9 Jars. During inoculation 2 jars slipped from my grip and tilted at least 90 degrees. Overall, I feel very good about the inoculation set up, and in a few days time I expect to start seeing healthy mycilial growth in at least 7/9 jars.
The following is my inoculation lab as it was used today:


To the beginners or even those who are just curiously reading, the total initial cost of this entire operation is less than 200$. If you live in a well equipped house or already have common household items like large plastic totes, pots and pans, pressure cooker, your start up costs can be as little as 50$. Going into this grow, I already had a SGFC, SAB, a drill and a Pressure Cooker. I purchased fresh jars for this grow at 11$ per 12 jars. Vermiculite runs 4$ a bag where one bag can get me through 30 jars or more, and 3 $4 bags of perlite suffice fine for adequately filling my SGFC. For purposes of anonymity, I can only say my Spore Syringe was purchased 100% legally from a vendor on the Internet for $19. I have also acquired additional blank steril syringes for use with my Liquid Culture (~2weeks).

Constructing liquid culture lids...Here is my version of the LC lid. The lid on the right (one with the white filter on top with the pinched tube) is complete, the other is ready to complete. The tube is a brass plumbing insert that has been gasketed to the lid via high temp silicon. The tubed is filled with polyfill for filtration and gas exchange and has been covered by a piece of tyvek under micropore tape. The inoculation port is just a drilled hole filled with high temp silicon to form a self sealing gasket.

Day 0: First Day of inoculations
I confess, I didn't have fine grain vermiculite. Upon inspection I found my verm to be super course. I processed it in a very clean totally dry food processor to achieve my desired grain. I have no doubts that this will be sufficient, but I do have questions concerning using the course grain. Can anyone tell me if it matters or not? In this case, I knew fine grain definitely works, so I was prompted to mill it.

Day 1- day 2/3 for inoculations.
This morning jars 10-18 were inoculated with my second unnamed species-strain of spore. Today's inoculation went by super fast, as I was well warmed up from yesterday. An inkling of doubt has nestled in my mind about my first round yesterday. During day 0's inoculation I did not recall to wipe the tops of the jars with isopropyl prior to injecting spore. I very much hope that not all of my jars are dirt as they were in a pressure cooker over night and the jar lids were covered with foil, but as I say...I worry.
None the less day 1 inoculations went off without a hitch and I am excited to finish up inoculations for this PF TEK grow tomorrow. I will make sure to post pictures as soon as mycelium is definitely present.
In other news, I have made the decision to include as part of this grow log my very first attemp at a bulk TEK grow. I plan on preparing a water/honey liquid culture (something I've done successfully several times before) and using that to inoculate at least 4 quart sized jars of rye berry grain. Preparation for the liquid culture is not scheduled until 8 days from now, after ALL of my PF TEK jars have had time to reveal their livelihood.

Day 2- Third round of inoculations: Unfortunately I was unable to secure the required jars last night and was thereby forced to run around this morning and acquire the final round of supplies for this Pf TEK grow. I have since yesterday decided to inoculate 36 half pint jars still staying with only nine jars of the unfamiliar strain. I have acquired my own preferred brand of fine grade vermiculite and I have to conclude that if you want things done correctly, always do it yourself. The latter 18 jars of this grow (now 36 jars total) will be prepared using the new vermiculite.
Because of the hold up gathering supplies I rushed to construct the lids, build the substrate and finally prep the jars for sterilization. It is these kinds of incidents that make me happy I invested in a pressure cooker on my first grow. Because I use PC sterilization in lieu of stock pot steam sterilization I am able to save some time, and I believe the nine jars I prepped and sterilized this afternoon will be cool enough for inoculation tonight.
Today I also started purchasing all required supplies to attempt a bulk grow from grain jars and a coir substrate. (I will start a new grow log for that grow when I prepare my liquid cultures in a week or so).
No previously inoculated jars show definitive signs of mycelial growth yet, but it has only been 48 hours for the oldest ones. Tomorrow I think will be when I start seeing something.

End of Day 2- As predicted, the jar I sterilized this afternoon in my PC were perfectly cool enough to inoculate by half past midnight. I feel much better about this round than any of the previous due mostly to the acquisition and employment of my preferred brand of vermiculite (no milling required). I will not be sterilizing the jars for round four of inoculations until Saturday night opting not to pressure cook them tonight or tomorrow morning. I am relieved that I already knocked out preparing 48 PF TEK jar lids.
At this pointing come to terms with needing to construct a third SGFC. Of course this means more perlite must be acquired. Putting the road map of this experiment together is proving more cumbersome each day but I am anxious to use this round of the PF TEK as a warm up for my first attempt at a bulk TEK grow.
I will post a new picture of my first sign of definitive mycilial growth as soon as it becomes evident. At this point I am confident its there. Happy cultivating, shoomerites, and I will be sure to log any on going progress.
it should be noted that inoculations were conducted while wearing clean long sleeved clothes as well as latex gloves that had been rinsed with isopropyl alcohol upon donning.

Day 3- I altered my schedule today by opting to commence the filling and sterilization of jars 28-36 this afternoon so that they can be inoculated tonight keeping a consistent schedule for incubation and consolidation.
The jars inoculated on Day 0 are now showing signs of mycelial growth at several inoculation points across the lot. Unfortunately at this moment no single point is broad enough to get a decent photo of, but they most likely will be by tomorrow morning if not late tonight.
It should be noted that for the first time ever I have not replaced the aluminum foil lids on the jars after inoculation. Indeed, as per the procedure in the Let's Grow Mushrooms videos I have been maximizing my investment by reusing them when able for each days new round of sterilization.

END OF DAY 3-
Jars 28-36 have been inoculated in what is easily my smoothest inoculation procedure to date. All in all I feel pretty positive about all 36 jars I inoculated over the last 4 days. I feel relieved to not have to prepare any more PF TEK jars for a while (ready to use my PC to make some chili)
As stated earlier, several of day 0's jars are now showing mycelial growth at several inoculation sites across the batch. I did take a picture so the very curious can see what to expect 1-3 days after a successful inoculation.

If you click the photo to zoom in you should be able to see the young mycelium.

Day 5-
While day zeros jars are all showing healthy mycelial growth, day 2 and 3's jars are all still blank right now. Several jars in day 1's batch show at least a spec of mycelium. The good news is that all inoculation points that are showing mycelium at this point have revealed no contams.
I have two isolated PF TEK jars that were not inoculated and not sterilized, they both show bacterial growth inside indicated by a distinct discoloration to the substrate, none of the inoculated, sterilized jars are showing any similar traits. At least right now I am confident That I have achieved yet another contaminant free grow.
it should be noted that until today (Day 5) I have been storing the jars in open air at room temperature. This afternoon I added a space heater to that environment to bring my jars closer to an external temp of 80 degrees F which should bring about an internal temperature of ~85 degrees F, an ideal incubation temperature for PF TEK jars.
More pictures coming when the colony u gets to a more formidable size.

Day 5-
Went a head and sed a shot of the most advanced colonies..,you should be able to zoom in for great detail.


For new cultivators and curious minds alike these pictures show examples of what to expect from a PF TEK jar 3-5 days post inoculation with a MS syringe. Inoculating with LC (liquid culture) will yield faster results where jars could be close 15-25% colonized after 5 days (as per my own experience with LC).
The space heater placed in the jar room has brought the room to 80 degrees F. I hypothesize that the addition of the heater will bring about a noticeable boost in colonization speed and I expect to start seeing new mycelium in all inoculation points previously lacking any signs of growth.

In the Lets grow Mushroom videos presented by RR, he explains that 70-75 (normal room temp) is okay then goes on to explain that the closer you can get your jars to 81 degrees the faster they will colonize. the link for the videos have been provided in a previous post on this thread:

The reason he says 81 is "perfect) is because that would make the internal jar temp ~86D F...the ideal temp for colonization.

Day 7-
As it was noted in this log, the two batches inoculated on days 2 and 3 respectively were prepared using my preferred brand of vermiculite, a very fine grain, I am annoyed to report that of all 18 of those jars maybe three of them show clear signs of mycelial grow where it has been more than 4 days for all of them. Meanwhile the jars containing the vermiculite I had to mill myself into. Finer grain are growing at a very good rate. Where some of those jars have quarter sized colonies going. It leads me to believer that a finer grain very might take a longer time to colonize initially? I don't really think so...but my results aren't what Im expecting today.
