WBS/LC experiment .
    #21328549 - 02/25/15 02:41 PM (9 years, 10 months ago)

I want to preface this by saying LC is not for everyone...if you are new to this hobby and are not perfectly comfortable with sterile techniques you could end up with a real big mess real quick. I would also like to keep the arguments about whether lc is worthwhile to a minimum please:-)

I seem to hit a wall with regards to myc growth when using honey. I can only get a small mass ( in relation to the amount of water) before the myc stalls do to running out of nutes( evidenced by the water being clear) so I want to experiment with wbs soak water in varying strengths to see if I can up the mass. I'll be doing the PC run tonight already have the wbs water in the fridge. I'm thinking I'll do one ten percent, one twenty five percent and one full strength. I know there's been quite a bit of stuff done with this but more documentation can never hurt.

I'm going to be using glass baby bottles and I'll be pcing a few syringes full of water in case they vacume (which happens pretty often for me. I like using the bottles because I just turn them upside down and the mass sinks right to the needle so I can get a concentrated syringe full of myc. I realize this method isn't ideal for lc but it is very functional as far as sterility and I just want to try and maximize its potential. For the record they will be nocced with a very aggressive cambo culture that's on its third or fourth transfer.

Anyone have any comments or recommendations?

Soak water is a well known method.

I use oat boil water for LCs.

Yes I know its well known but I want to see what percentages work best for bottle tek LC which I haven't seen specifically tested with any kind of control...and certainly not tested and documented enough to have a clear answer on what percentage creates the most mass per volume. I plan on doing quite a few variants ...just want to use this thread to document my findings.

I'm mostly a lurker but figured I might as well contribute lol. I've been shut down for almost a year after a soul wrentching trich outbreak. But I decided it was time to roll out the tubs again lol....fuck trich!!! Anyways nice to meet ya:-)

Ended up doing one at 15% one at 30% and one at 60% For the fifteen and thirty percent bottles I used wbs soak water that was drawn off the top of a quart I had in my refrigerator....for the sixty percent bottle I drew from the bottom and got some very fine sediment as well as a slightly darker liquid. The wbs water hadn't separated nearly as much as I had expected it wouldhave as it sat still for a week prior to use.

All three were inoculated with an agar chunk from a cambo twice transferred plate. The plate was showing extremely vigorous growth so I expect the growth to be pretty fast in the nutrient rich mixtures...hopefully creating a more significant mass than honey was creating.

Each bottle has a flint arrowhead in it to help with breaking the myc up and the bottles will be shaken twice daily.

I have 2 full strength qt jars made with MS in less than sterile conditions. Has been over a month. I'm tempted to strain one knock up some plates and then shack the other one and make a syringe to test on plates too. I'm a bit curious to see what I have been growing. Looks cool.

What percentage of the liquid has the myc colonized?  A substantial amount? I'd really like to get my percentage up as high as possible. Using honey in the bottles I'm getting like 10% mass at most . I'm sure I could increase it some by using a stir plate but im simply looking to maximize the bottle tek because I like having the closed environment it provides.

I've used salsa jars (with poly stuffed syringes as filters)with pretty good success but don't like having to open them up to draw out the myc. I know there are way better ways to do lc but if I can up my mass even to double what I'm getting now it would be ideal for my use.

OK so the 15% jar did fair , maybe fifteen percent mass( and has stalled). The thirty percent is still going strong and it has reached maybe fifty percent mass!!! The 100% is still going strong and is looking like a damn blob of myc!

I nocced some grain jars with the thirty and after 72 hours they are looking great. I also nocced several little pucks of brf mix . To be used as slurry. The brf mix showed significant growth at 14 hours!!! I've never personally seen a recovery that quick. Although I never use brf. The recovery and quick ness really surprised me. I'm impressed!

This experiment blew honey water out of the picture for me. WBS doak water is vastly superior in at least as far as baby bottle tek goes. I think I'll be using a fourty percent solution from here on just because you can see through it good and the myc mass will be ridiculous!

If you're new to lc and want to give it a whirl I'd say this would be a great start.
