My SHIP are made of silicone (like many people i guess), so it doesn't really matter if i have to redo it after 2 or 3 uses

Correct, the tube goes into the LC.
The tube is held in place by a media vessel-to-tube holder device.
Tube holder sold separately.





Had to shell out the extra cash for the tube holder though.
Would rather go with than without on the tube holder.
Otherwise, it was projected that tasking with gun would be even clumsier than already anticipated.

My stainless steel LC/I gun came in today.



The tube runs from the gun to the needle.
The needle goes into the media.
The gun has an inline pump action that pumps liquid into the glass barrel as liquid is aspired.
When the device is empty, it takes a few pump actions until the device and tube become full of solution.
The culture stock to test out gun is being generated (ATL#7's)



200, 2 foot tall, large gusseted spawn bags.



That'll last me a month.
Maybe two.

The plunger leaks on disposables after several PC runs.
Glass syringe has glass plunger... Or atleast looks like it from the pic :/ mine will be here in the mail from overseas in a day or so. 12.99 shipped! order from the states same size, 50 bucks.

I've never had a plunger leak, the glass ones, not having a rubber plunger have zero ability to push out contam spores that may be on the inside of the barrel during aspiration, and if you pull out the plunger too far, the airtight seal is broken, contam vector city.  Also they are fragile at the plunger head, the glass is very thin there and not hard to break.  Plastic is my pick.

And if your pc seal gives out half way through a cycle, you loose pressure fast, which can cause the glass to weaken and or crack.

Nice! Let us know how they work out! I'd like to see how much difference there is between the metal one and the plastic one .

I'd think that putting the tube through a metal lid and siliconing it on would be way more reassuring for sterility... I'm assuming the tube is detachable from the gun.

When making your design, don't make it so the tube hangs into the LC during colonization.  The lc will grow in the tube and clog it.  The gun will not have enough suction power to overcome the lc-plug in the tube, and even if it could, it would clog the syringe.

Here's a quick look at what I figured out.



Design was inspired by the example photo in the instruction booklet.
The same design can be replicated with metal lids and silicone as you described above.

I go to the absolute extreme when it comes to sterility on LC so keep that in mind with the following comment:

Those plastic lids can introduce foreign spores by means of the space between the lid and the glass rim, unless you have a decent gasket.  That lid will probably work 99%, but I've seen them fail in just the way I described.  To put it another way, air currents can cross the barrier between the lid and the glass rim no matter how tight the lid is cranked down.

No gaskets are used.
Instead parafilm is triple wrapped around the space between jar lid and glass.
Mediums are left inside the vacuum of the AA until inoculation.
LC lids are removed for inoculation in front of a flowhood in an enclosed lab.
Then the parafilm is wrapped onto the media vessel.
Directly in the downstream of the laminar flowhood.
The mediums are then placed on a shelf and incubated at 70-75F & 40-55RH.

Contams travel in air currents.
Air currents will take the path of least resistance.
These principles are crucial as to why it's critical that a GE filter be placed on an LC media vessel closure.
So that the path of least resistance for air to enter your media is through a media filtration device.
