Contamination ID Please
    #21355194 - 03/03/15 07:11 AM (9 years, 10 months ago)

Hey guys, two of my PE mini-monos contaminated. The colonization stalled out on the CVG (bucket tek :uhoh:).

I was worried about these tubs as they took so long to colonize. Initially, I was fearing the Sterlite latchbox lids were not allowing enough GE, but not sure if that is really the issue (will need to test an isolate on two different tub styles to confirm.

Thoughts on what these contams are? They are both neon colored (you can see green and red).

It's either a type of Aspergillus or it's Serpula lacrymans. Toss that shit

Yeah I tossed it the minute I saw the spots. First PE tubs :frown:

Ah well, I started properly pasteurizing my substrates even though peeps say it isn't necessary with CVG, still makes me feel more secure.

Yea man, always pasteurize properly, it gives the myc a good head start to take over the sub before mold can even think about it. The best easiest way to Pasteurize is boil a big pot of water and fill a cooler about 1/4 - 1/3 full of boiling water, put your mixed field capacity bulk in gallon Ziploc bags about 1/2 full and throw them in the cooler and shut the lid, wait 3 hrs, pull em out, dry the bags off and let it cool down for an hour or 2 until it feels warm on the face. Then it's ready to go. Been doing this me this method for a while with no issues

What type of grains do you use? What temp do you grow at? What type of light do you use?

The coir/verm/gypsum bucket processing may not have been the issue.

Are you sure your spawn was clean?

I'm pretty positive the spawn was clean. Autoclaved at 120C, inoculated in a flow hood with a syringe from a trusted source. I don't believe the syringe was contaminated since I did put some of the syringe on agar plates too and everything grew out clean.

I use rye berries, grow around 70-73F, and during colonization of these tubs I had no light source on them due to my daylight bulb lamp being used on fruiting tubs.

I should mention the grains were fully colonized for about a week before I spawned them do to my schedule. Perhaps that explains slower colonization on the substrate?

It's possible, but I doubt it. I've used some multi-month old spawn with no issue before.

Perhaps the contamination was the cause of slow colonization? Maybe the slow colonization allowed the contamination time to set up show? Hard to say one way or the other, hopefully it's a freak occurrence.

Yeah I definitely hope so, know soon

Cheers!
