Ive tried this twice. Two drops on the agar and nothing after 2 weeks. no visable contams. no myc. I used the recipe on page 1. After failing i didnt worry about it too much because I was super busy with all my g2g. BUT I need to get this down so I can move to some pe strains.

I had drops all over the sides of my plates after pcing. I store them in a closet after knocking em up.

I have yet to even inoculate, (probably tonight), but from what I've read spore solution is kind of hit and miss on agar. :shrug:  Again this is just something I have read. No experience with it yet. Maybe someone more experienced can chime in. In multiple threads I have even read of TC's having bad luck with it.

Yes ms solution sometimes can be hit or miss for me at least. Every now and then I just come up with no growth. I know many people say they get bacteria but for me its just a blank. Usually when I am using ms solution I do up 4 plates and then if more than one gets growth I transfer from the best ones and forget the rest.

I have also read that MEA is better for spore germination, and PDA is better for clones and the like. I am trying to find the threads I found this in to link them. I forgot the keywords I searched.

Anyone experience this or wanna chime in?

No I must disagree. More nutes and softer agar is better for germination. I used PDA to germinate spores dozens of times. They both work, people develop preferences of course but really you should get in the habit of rotating at least 3 types of media if you intend to keep your cultures long term. So just a quick aside, I use MEA in most of my slants so I almost never use it in plates that will be receiving or transferring back into a slant.

Hmm where you hear that?

Both can be good for spore germination as with both you can lower their agar agar amounts making them more aqueous.

Simply because it's just better to rotate foods. Would you like to eat always the same? Same with grains, g2g from wheat to oats, etc.

Im looking for the threads now. I think I searched something like 'spore syringe to agar'. Something along those lines. I kind of had the thought that softer agar would help germinate, no matter the recipe. Im no expert and just wanted to bring it up.

Softer agar=better spore germ, would be more fair to say than MEA is better than PDA for germination?

Basically what Alien said. Rotation of media helps maintain vigor and prevents the culture from becoming lazy. After chewing on the same food source in a slant for a year or two changing media helps it bounce back better with fewer transfers. This means a better state of vigor when I transfer it back to the slant as well as when it goes to grain or LC.

Just bought some ingredients to whip up a few of these.

I got potato powder instead of flakes, i'm assuming it's same diff since it's all potato starch basically. I only eat raw honey already so I have that and I got good old telephone agar. I forgot the food coloring though, clear agar for me.

When I first read the tek I actually thought of the larger sized, shallow ziploc or glad containers that are about 4 or maybe 5 inches across and maybe 1 and a half ish tall. I didn't see em at walmart. anyone used those to make plates? the bottoms were nice and flat as I recall. if you wanted to make divided plates they'd be better too since they're bigger (saw someone mention that when I was reading through the thread some, thought i'd say something)

I'm fucking pumped to get into agar and then start trying some LI. of all the cultivating methods i've read about so far long term the one i want to pursue first is definitely agar>LI>grain>mono. not sure what grain yet but whole oats look neat and i'll probably use coir a lot but i also would like to try straw.

i'll probably try to whip some plates up on friday, and have them ready to take some pins when my mono starts pinning and some clones from a few choice looking fruits. gods be willing.:sun:

I've used the larger round ones.  They worked fine.  The only issue I had was I bought some really of brand ones, and after the pc cycle the lid was a bit looser fitting than when it was before. It still worked out though.

Next time I'll have to hunt around a bit, and see ifi can find any interesting pp5 containers to mess with.

These are pp5. Got six of them from dollar tree. I'm going to test them out when I finish my flow hood. I was thinking about taking the handle off and putting on silicone to seal up the holes in the lid.
Each circular slot  holds about 30 ml of liquid. I will have to pour the agar because the container has to be sterilized on it's side. It will not fit in the pc in the upright position.
