Traits for selection

When isolating strains of p. cubensis, which are the traits to look out for? I am eager to experiment with isolating true breeds, but lack a good overview of the traits of this mushroom.

Thanks, etc. I am beginning to get a decent grip on agar work. My knowledge of mycobiology is adequate, that is I know a little about the life cycle of basidomycota, mono-/dikaryon hyphae and meoisis in fruiting bodies. I've been reading up on here on strategies for isolating true breeds. What I am mostly wondering is what traits to select for visually.

basically what shroomydoo22 said. Recently in search of a strong culture I made two different agar recipes (lme or potato), each transfer I switch mediums so that which ever growth is fastest after a handful of transfers I can be pretty sure is a strong colonizer.

you said visually I assume this means on agar. I think it's worth mentioning that cloning young pins (can be done off agar) can capture genetics that are vigorous in growth and the important part: almost certainly a good fruiter. where as from spores if fastest growth is selected it may turn out that the result is not the best fruiter but just fast (who wants that?). Always remember to do trials, never all eggs in one basket.

Without fruiting each isolate out there is only one trait that can be selected and that's strong healthy colonization. Otherwise you will need to fruit them to see what you have. Fruit them all and keep the best. Usually one in ten is really good.

But dont forget potency! I prefer to clone from mature clusters so I can save the mushrooms and eat them when I get a chance, i can get an idea of the potency while Im growing out the cultures

By young pins I mean like the first picture in my user gallery (can't find how to paste on mobile sorry). I mean the first very small mushrooms you see start to form. Some look for clusters of mushrooms to clone from to have a better chance at producing clusters from the clone.

As apposed to cloning a mature fruit with its cap open.

my only contribution to this thread is to stress the important of proper labels.With out them it makes no difference if you took a good isolate or not.

Great point too. It gets confusing if you don't label well.

I always put the date, "strain", and generation. I just start with Spores -> Agar -> "PE" 1, "PE" 2, ... PE1 -> PE1-1, PE1-2, etc...

If that makes sense haha. Helps me stay organized on the plate and sectors I transferred from.
