hello all, 4 days ago I nocced up 16 pint jars of rye grain spawn and I have been checking on them casually day to day, I expected to see my first signs of growth on the surface of the grain because that's where the spore solution was injected but I noticed on the very bottom of one jar that maybe 10%-15% colonized by light, fluffy but pure white mycelium. I highly doubt that there could be any kind of contam on the bottom of my jars because I pc'ed the dickens out of them, but I do have a theory I would like to share with all of you, see what the community thinks.

prior to being pc'ed my grain was a little too wet, it had soaked for close to 48 hours in warm water and a bit more of the grains were exploded  than optimal but it turned out okay, just a little too wet, after they cooled I shook the jars to break up the grain that clumped together in the pc and the grains at the bottoms of the jars got kinda smooshed to the jars bottom, and remained kinda pasty, after the inoculation I put them in my incubator with an aquarium heater but I think I set the thermostat a little too high (mid to upper 80's) on the heater because after about 2 days I noticed that where as before the grain was too wet they now seemed I little dry with condensation on inside of jar in the space above grain.

now that I see this growth on the bottom of my jar I theorize that perhaps I am experiencing some kind of moisture reflux in which the heat from the incubator caused moisture to evap from the grain, re condense at the top of jar and run down the wall of jar to the bottom, taking with it my spore solution, im concerned that the spores will not enjoy the environment at the bottom of the jars because of excess moisture and was considering turning them on their sides to redistribute moisture. I don't want to shake them thought for fear of hindering any early mycelium that is forming

has anyone ever experienced this?

update: 3/2/15

so im exited to add that every single jar shows some degree of healthy growth today at the 7 day mark, and so far only one shows any contam, it was my syringe wash jar so I fully expected it to contam, it has a patch of green mold that encompasses literally a single grain on the surface at the moment, other than that out of 16 jars it breaks down to this:

:16/16 show mycelium growth
:1/16 show contam growth
:5/16 are approx. 1/4 colonized
:1/16 shows patches of strong rhizo mycelium.

Possible explanation. I wouldn't recommend an incubator. You don't need it, and higher temps are going to increase risk for contamination.

yes I realize I soaked my grain for too long, its been a learning experience getting the moisture content just right, the first batch I tried to simmer but it turned out much too dry and had to be discarded and this batch too wet with the soak, however I feel I have learned from my failures and the next batch should be just right,

as for the growth ive seen, after I typed out my post I went back and looked over each jar carefully, and I must say it did much to alleviate my fears, quite honestly I think it is far too early to say anything at all but it looks like healthy mycelium to me, im going to give it a few more days them upload some pics for all to see, taking pics now would be pointless, to young to tell, its just my paranoia, making a mountain out of a molehill, jumping the gun, ect. ect.,

ill keep this thread alive as my grow log though, I had planned to start one anyway once I saw some growth, and I guess technically I did, lol

Not sure why you allowed your grain to soak for 2 days? Also most peeps do quarts of grain. I read you are doing pints? Any particular reason for pints? Or just what you had on hand? Depending on how much spore solution you injected in each pint. Spores could have easily washed to the bottom of a pint and started to colonize the bottom first. Pics of the jars are a lot easier than guessing though. :super:

Welcome to the Shroomery! :mushroom2:

Sounds like you're learning the same way everyone else here does, by trial and error and making mistakes and learning from them. That said, it's highly possible you've found out one major issue in over hydrated grains, it's a huge vector for bacteria.

And yes, resist your urge to shake the jars at this early stage, it will likely ruin your project.

Keep us posted and ask questions. Everyone was new at one point in this hobby.

thanks for the responses,

yes, I used pint jars because it was what I had on hand, and I used about 3 cc of spore solution per jar in a single injection site

also now that I think about it, it wasn't a full 48 hours, much closer to 36 and I had intended to pull it earlier but was delayed getting home from picking up mail at the post office,

upon closer examination I noticed very early growth on several other jars as well, I hoping I have some good pics to show the community by the 7 day mark,

although initially I took this growth as an early contam scare I now am very optimistic about this grows future, im hoping it will have a higher success rate than my previous attempt at grain spawn, (40% w/ pre-sterilized grain bags from out-grow) will update when new development arises.

day 11 update,

okay sorry guys I meant to update this yesterday but I was unbelievably stoned and forgot, so the growth I saw between days 7 and 10 fell a little short of my expectation and I was worried that I might not see full colonization by day 14, on the morning of day 10 I checked in on my jars and noticed that they all average between 1/4 and 1/2 colonized so I figured it was time for a shake, so I shook and rolled all my jars to break up and redistribute the mycelium and put them back in the TiT incubator.

at the time I was concerned after I shook them because they didn't look very good directly after the shake, I felt like I might have just killed them.

now I just looked at them again at the approx. 36 hour mark since the shake and all such fears are alleviated, they look awesome! i'll take some new pics tomorrow and upload them but so far they look like they have recovered very quickly and are primed to take off, my faith in a 14 colonization has be recovered!
