Noob Idea, thoughts?
    #21333739 - 02/26/15 03:40 PM (9 years, 10 months ago)

First off, I did check the search function and didn't find info on this.

What would happen if you prepared a liquid culture jar with a piece of cheesecloth fitted under the lid.  Then rather than taking spore print and transfering it, you literally just set a cap onto the cheese cloth to drop its spores directly into the jar?  Inside a SAB of course.

The thought is that any contaminants that might fall into the jar would fall into a spore print anyway so why not skip the extra steps involved in transferring spores off a a spore print into a spore syringe then injecting it to a jar?

Am I crazy?  If so, why?  Trying to learn.

Because any contams that fall directly into a nutrient rich solution will multiply extremely fast. 1 contam = lost LC  where as a spore contam on a print is only in one tiny spot and not able to reproduce until you finally pick it up.

Thanks for the quick response, to play devils advocate wouldn't you just pick the contaminant up off the spore print later anyway?

yes but you use a very limited amount of spores when making a syringe or going to agar so you will get 99 good innoculations before you get 1 bad one.

you would end up with a jar of mold since a LC needs to be inoculated with perfectly clean medium hence why the only right way to start a LC is with agar. if you're not making a LC with agar you're doing it wrong.

also it would be way too many spores. less is more with spores.

and yes spore prints have some contaminations on them but they don't necessarily matter since you use a small amount of spore solution to inoculate and they may not germinate on PF cakes and if you use agar and they do grow then you can separate clean mycelium away from the contamination. in a liquid nutrient like LC they would just grow and fuck the whole thing. LC is a step backwards for most new growers as it adds another step and time onto your grow.

I'm not sure what exactly you mean here, if your going to do a lc and add spores, one with 100 spores and one with 10,000,
would you say 10,000 is "too many".  I'm kind of getting hung up up on the phrase "too many" akaif, there is nothing wrong with having a lot of spores, if they are clean.

I think what your really intending to say is that considering a print/syringe with even a small amount of contams, less is better to avoid the contam issue, but the actual number of spores does not cause a problem even if the number is very large.
Do we agree here or is there something about large numbers of spores...?

too many spores too many genetics, you will have so many incompatible strains on your fruiting substrate that each of them will be fruiting from a tiny chunk of substrate. so in that way yes 100 would be better than 10000 spores in an LC

I thought that mycelial networks knitted themselves together to work in unison to form a single mushroom, don't stem clones usually
carry several sets of genes necessitating the need for further sectoring before an isolate can be taken from a clone?

the whole thing isn't competing with each other it's acting as a unified mass of cubensis. but yes more than one dikaryotic strain can meet up with another dikaryotic strain and make a fruit body itself that has got multiple genetic strains. hence why some clones are not an isolate right way. there's also dikaryotic strains that are just not compatible. the less strains the better be it cakes or bulk or LC or anything between
