Agar sectors

Hey everyone how iz ya?  I myself am feeling pretty good.  I just finished catching up my grow logs and doing a little bit of mycology work too.  Now I'm working on a nice beer buzz and I have been :flyhigh: all day.  One of the projects that I am wrapped up in right now is I am working with agar trying to isolate 3 separate cube strains.  I am going to make some transfers tomorrow.  I would love it if you guys and girls would take a look at these cultures and help me figure out where the best places to pull from might be.

I'm thinking 11 o'clock again.

Now I am basing my sector areas on how good the growth looks and how easy it is to define as far as an obvious sector would be.  Please let me know what you think the best areas to pull from would be and why it would be.

Your labeling says, "solution", so I'm thinking you dropped spore solution on the agar.?

Lots going on on your plates.

Grab something clean from the leading edge of your growth and transfer that and you'll reduce/simplify what you got there.

one point of origin makes things easier.

You need to look through your plates to see where the actual mycelium looks the best.

One sector may look good on top, but not have any rhizomorphs when viewed throughout.

I think I'm currently on plate 9 or 10 with my xapuri, and I'm just starting to see hectoring now. Sorry I can't post pics currently

So those were the germination plates? First plates?

I don't know, I've yet to grow Pans.  I am going to give them a go some time soon.  I want to get my feet wet with panaeolus cinctulus then once I have played around a bit and gotten used to the spieces I am going to give it a go with my panaeolus cambodginiensis prints.  I am actively looking for a wild panaeolus cinctulus to print so I can start that project up. 

As far as the wrap, thanks man.  I was having troubles with my parafilm at first.  It wouldn't stretch too well and it was brittle too.  I think it might have been because of the colder weather because it has recently heated up a bit in my area and I am not having any problems with the stuff anymore.  Each dish gets 4 squares split in half.  I can get this to wrap around each dish 2x.  This way I am able to double wrap 2 dishes with 4 parafilm squares.

I use LME.  The agar is thin, but not super thin.

mushpunx, that plate is the 2nd transfer.  I shot a vender syringe into a grain jar (I knew it was risky) and it grew mycelia and bacteria.  I took one rye kernel and put it on agar

and then transferred a clean piece to new agar

rye to agar to agar.  would we call that 2 transfers or 3?

Yea id call it 2nd transfer. I asked cause it looks like it wants to show sectoring already

I love putting a kernel on agar, sometimes thats the easiest way to get germination
