Incubation in 2 litre plastic bottles?
    #21312019 - 02/22/15 08:46 AM (9 years, 10 months ago)

Hello.

Question!

I have 24 jars, 12 of which are almost fully colonized rye...

I want to flower only a couple and then use the rest to spawn more...

Now since I only have 12 or so more jars left over, is it possible to pressure cook the rye in those 12 jars, then dump all of that into a clean 2 litre pop bottle and spawn that in my incubator?

Then I could just keep pressure cooking in the jars and dumping into 2 litres. Therefore not having to purchase more jars, and be able to colonize a big 2 litre...or is there something wrong about colonizing in plastic?

I'm not an expert, so take it with a grain of salt.

I don't know of any reason that it couldn't work. The problem is those 2 liters are never going to be anywhere approaching clean or sterile. Rates of success are probably going to be very low.

I never did work with bulk spawn though.

Yeah, I was thinking there might be something wrong with it because noone(online(that I can find))is doing it... but if the bottle and everything is kept sterile.. I'll try..

Oh, and this is kind've how im going to have my terrarium...
Except I'm going to have the wick run into perlite, and attach another bottle nozzle on top, with a hole poked in the lid and a filter...
Then it will do all the work for me, stay moist and not need burping,

That is the in vitro plan right now, wash the cake and flower it in there...

I think it will be just enough space they won't squish too much...

sounds like an incredibly good way to grow contamination, you're over thinking this soooo much, they're not plants. plants rely heavily on bacteria in the soil. you run a wick setup through perlite and then with the lack of airflow you're going to have a gross mold factory.

if you want to do in-vitro just do in-vitro then. make a pf cake in a larger jar, skip the dry verm barrier, use a grain jar lid. and fruit it with the lid still on.

Jars are pretty cheap. Think of the time and material wasted on high rates of contam vs just buying another dozen jars.

Personally, I'd just buy more jars. Unless your only goal is to experiment, it's just not really worth it.

But like I said, I'm no expert. I only ever did PF Tek.

I'm still going to try it though with one of my patties now... bod-totally right about over thinking it I'm on a coffee bender and spent all night cruising here and plant sites...

Yeah more jars is the way to go.. thanks guys, I should just stick to the normal methods and then just experiment a little at a time

I guess I kind've put 2 unrelated posts in here, well related by the 2Litre pop, but 2 different theories...

mushrooms should be the only organism present. using plant watering methods are not proven methods to keep environmental pressure off of keeping other microbes. they in fact encourage it because plants need microbe activity. we want only the mushroom mycelium there and want to give it the best chance possible. open watering reservoirs need to be dumped regularly. most of the time if we want to dunk a substrate like a PF cake we can do so in a dish of water for a few hours. it's not perpetual, we don't leave the substrate or cake in water for more than the few hours or it risks getting stagnant and then nasty.

Just say someone was doing bulk PF cakes. Would they still want to dunk the cakes?

you mean big cakes? or using the cakes as spawn for bulk substrate?
either way if you're making a big cake I would just water it and pour off the excess after a few hours. if you're using it as spawn you can dunk then shred it up for spawn similarly to dunking grains before spawning.
