First time wedge use
    #21295465 - 02/18/15 08:06 PM (9 years, 10 months ago)

I received a wedge and the little vial it came in opened in the process of being mailed to me. I needed to make some agar dishes quickly in case it were to dry out. Now I thought to myself maybe I should make the best out of this chance. So I got everything ready and pLaced them in my SAB. I sterilized the knife I used and sliced the wedge into 3 pieces and placed them onto the agar. All 3 of them seem to be starting to show signs of life. I was looking for illustrated logs of wedge to agar so I would know what I should see and shouldn't see. I wasn't lucky enough to come across what exactly I was looking for. So let me pick your brains plz..

What should I be looking for?

What's the next step?

Did I screw up by cutting it up?

Thanks

You should be looking for healthy growth coming from the wedge.  You definitely did NOT screw it up by cutting it with your sterilized blade, and placing a piece of it to several plates was a good idea.  If this was a culture sent to you, chances are pretty high that it was a decent one, so trying to isolate would be a mistake.  Just wait until it's spread enough that you can transfer to make absolutely sure it's clean, and then transfer from the second plate into a jar of grain.

Thanks for the quick response was kinda nervous I may have made a mistake.

When looking for healthy growth...is it supposed to look like it has white fuzz growing around it?

Yep.  Check out Stro's cloning and isolation tek for a lot of pics on what it should look like.

Yeah, Stro writes a really good tek.  I wish he came around more.

A lot of people call that stage "leaping off," FYI.

that was the right thing to do..
are you wrong.. of fucking course not!

idiot customs people like to shove everything up their asses for no reason and ruin cultures all the time.. that's why I order spore prints from seed rings.. cheap and safe.

hopefully its already isolated.. which will mean that it will all grow as a single circle evenly .. check RR's videos about agar.

in your case you might just need to isolate it from whatever else grows on the agar... think the mailman's anal flora..

onto a clean jar then onto an LC and substrate that you will G2G for ideal results (Y)

happy shrooming

If they grow correctly with no contams would it make sense to make an LC? I would like to possibly make some syringes out of it

agar is the best way to make an LC, so yes.
but i would also consider dropping some wedges into grain(s) as well for backup in case the LC fails.

I was think the as well. If possible use one of the 3 for LC and another for grain. The third I would put on a master slant to preserve for future use.

I second this idea.

I've been very impressed with my recent agar wedge into grain growth rates.

a seed ring is a place the helps circulate mushroom prints for a small fee unlike commercial sellers. its done in a way to spread the "magic and divinity" behind the psychedelic experiences without financial gain..
at least that's what I understand..

here are ones that are tried and tested

http://fsre.nl/

http://freespore.com/index.html

they are rather slow though... but they do work.. also the risk is very little.. so don't think about them much.. but you will be happy when their prints show up

LC any time,

Grain I'd wait for it to get a little bigger then that.

If transferring to lc should I just take a small wedge of new growth and leave the original wedge to continue to grow?

So I did a transfer to a rye jar. I decided to do the one jar to see how it takes. If it works I will do g2g instead of putting wedges in all the jars and risking it in case of contams.

I figured that was the case. So I I will wait for this to colonize and then G2G. Maybe even do a small container of grain that will colonize quicker then a big jar.

I only worry I will have so much substrate colonized and now know what to do with it. I have only done cakes and a small make shift casing.

Cool.  You can really see where your mycelium is growing up and over the bacteria there.

I was going to ask about that too. I assume those strange circles out by the edges are some type of bacteria. Now do I let it continue to grow or transfer?

I'd transfer from a clean place.  Bacteria at the edges implies you're not getting a good seal, though.  Are you using the same type of containers?
