Look at this travesty...should I try to isolate away from the mold or just toss them out? *PIC*

So this was my second transfer away from the original MS inoculation and I got hit with this. My previous dishes were fine but I must have gotten lazy here or something. Should I try to take a wedge from near the center and transfer to another dish or should I just toss them and start over?

Haha... looks like the tissue was clean but your air wasn't... You could easily isolate away from that. Just be careful though because the mold is already sporulated. I would just pick that top center dish and leave the rest alone. That one looks healthy & has the least mold

I don't think I waited long enough for the air to settle in the still air box...what a waste of 20 dishes lol

Yeah first I wipe down the SAB with a lysol wipe then spray the inside with a solution of warm water, soap and a splash of bleach and wait 10-15 minutes for the air to settle...I wear gloves, tyvek sleeves and two masks.

I wouldn't worry about masks since its in a box. keep your bleach/water mix bottle inside the box and spray a mist into the air immediately prior.

My guess is those spores were stuck to your gloves and fell onto the plates while you making the transfers. Gotta watch what you touch after you put the glove on.

I usually "wash" them with alcohol every time I take my hands out of the SAB but I think I made a couple of the transfers after they dried, either that or the alcohol just wasn't enough and I need to stop reusing gloves.

Alcohol kills a lot of bacteria but it doesn't kill mold spores. Also you don't need to open the dish all the way. Just lift it open on one side, hold your tool(scalpel most likely) from as far back on the handle as possible, reach in with in with it and drop the piece on there. The hand holding the tool shouldn't be above the agar, only the end of the tool transferring the wedge should.

I took a year off and my sterile technique is awfully rusty...thanks for that.
Also, I've heard you can sterilize tyvek sleeves in a PC, how does one go about doing this? Do I put them in a closed jar and PC them that way? I'd imagine you'd want them as dry as possible

I've never gone through the trouble of actually sterilizing sleeves but yeah that should work. Otherwise you can just disinfect them like you would your gloves.
