if no one replies, you might wanna post it in the advanced cultiation forum, they talk of agar and address agar alot more in there

Well technically you could transfer at any time, but it is traditional to wait for the whole plate to colonize that way you have the chance to transfer more slices to more plates. Slice it like a pizza, slices don't need to be very large at all, like a square cm is pretty good. I would wait until the whole plate is colonized unless there is ab obvious danger of spreading contamination. Both mycelium and original tissue can be transfered, but you should transfer mycelial growth because it's somewhat redundant to transfer original tissue.
