Sporeprint question
    #2125341 - 11/20/03 04:51 PM (13 years, 6 months ago)

After a recent trip finding some azures near Astoria I took them home and left them sitting on a desk in my room for a few hours. I then decided to take some prints. I took a new piece of typing paper and layed the caps down then covered with a bowl. After letting them sit overnight I took the piece of paper and put it in a new ziplock bag. Is there a high chance that these prints will have contams? What is the best way to take prints of wild mushrooms to make sure they stay sterile?

Theres only ones way to find out, give em' a try, the best way I can think of is to bring a sterile jar with you when you go hunting then set the cap in there, then when you are ready just spray your water into the jar to make your syringe.

I have picked several wild shrooms and just put them in a paper bag untill i am ready to tke the print.
I just wipe a piece of foil w/alcohol and let dry. I put the cap on the foil with another piece of foil on top. Let it sit overnite and BAM print.

It was and will never be sterile.

To use wild prints, you have to do agar work, usually antibiotics are best, but you can sometimes work without them, using some tricks.

Don't worry about keeping it sterile though, it wasn't and wont be.

What kind of tricks do you use? I have contamination problems with my wild prints/clones ( mostly bacteria and yeast I guess ) and I wonder how a more experienced mushroom grower deals with this kind of problems.

When making a print from a wild specimen, and the spore load is heavy enough, you can discard the 1st or 1st couple of prints. The later ones tend to be cleaner.

And when cloning or germinating spores, using antibiotic plates (gentamicin or chloramphenicol) will keep bacteria down. As for other fungi, you just have to subculture away from them.
