Idk how I did it, but I must have had some spores on my hand or something, they landed in a line on the plate lol.

Not if u want penicillin.
So what does it tell u if the myc grow to fluffy and tooo agressive? Im not taking back what I said abbout Grain water because its the bomb for sprint start. Whats the difference between that mix and the mix u told me Pasty 10/8/2 A P D. How would I adjust PDA to get something in between.
I think I may use AW for transfers when u want to get far down the line quick.
Id like Violet to pipe in here as well seeing its from here advice.
It would be good to tick off another thing in this subject of the thousands of things I dont know......
Any to be honest my mates think im a microbio genious...if only they knew...

I use grain water all the time cause of its high nutes. It really comes down to your culture and what it wants. Then provide that. Some culture do better with less nutes some want more. Some prefer certain media to others. I have a figi that grows apeshit on PDA but terrible on MEA. You pretty much need to experiment as much as you can to find what works best for you.
I will say that so far grain water really does good for most of my stuff. My agaricus did poorly on it tho so I am gonna try different grains for the soak water to ensure good colonization before I attempt to make spawn with it. Gotta try lots of different things to really find what works good.

let me ammend this my agar was very hard There was definite myc but on cutting it was actually hard feeling under the knife blade. I went ahead with the transfer but not sure if it will take? Actually both plates seemed clean for what the prints were exposed to I think i'm gonna try a new batch of agar with more liquid this time. More of a jello like consistensy.

hey Pasty, u rock, love this tek...
love my mrs aswell... haha
She suggested, instead of using paper towel before wrapping in foil, she gave me some of those round make up removing pads. you know the round ones?
i used 5 of them on each and it eliminated all moisture... the are very absorbant

Will agar soften up if you add a little bit of water after it 's been pcd and set? My first batch of plates came out hard like cured silicon. Ijust threw all the plates out other than the 2 I had already transfered to. I still have the original and it hasn't contamed and colonised fully. The transfers seem to be lagging a bit.

The tyvek filters are working great once I used the drill bit to boar out a large oval shaped hole.
That perma filter.

This tek inspired me to get into agar. I wound up doing in it a bit different but I get results. I have made a few observations regrading agar that I thought I'd share. What is up with red agar having the color sucked out of it? Every cup I've run with red dye has turned clear over time.
I haven't used a PC for any of my agar and I have been getting about 60-70% contam free runs. Still room for improving there on my end. Mostly has to do with how I do it I think. My process is cheap enough to toss a few and steam roll along.
Its probably my imagination but blue seemed to have the healthiest looking stuff but probably a contrast thing.
Green seems cursed to attract contams. Again probably my imagination.
Tl/Dr- agar is a great and can be a very cheap way to start culturing. Thanks for the awesome tek/inspiration.

Here is how I modified these pasty plates with reusable tyvek from a envelope.

So has anyone else tried the cotton rounds instead of the paper towels? I tried them the other night, and yes they're absorbent, but they got stuck to the micropore tape. Left little cotton fro's behind. Maybe I got cheap ones?
