That's an interesting paper, I've heard about other uses for peroxide but not specifically for giving a competitive advantage when germinating spores. It's a shame it doesn't give more detail about the method they used to wash the spores though.

This is what they have antibiotic agar for.

Peroxide can have a very negative effect on your cultures.

Just saying as far I know antibiotic agar has no negative effect on growth. Last thing you want to slow mycelium growth when you need it to be growing as fast as possible so you can isolate it.

What is elses opinion:

Woud you use lower concentration of peroxide with cube, serbica or pan spores or you would shorten the time?

I will try to clean up serbica spores from dry fruitbody by washing and (soon as I get gentamicin) streaking on antibiotic agar.

Water won't flow through a 5 micron hole. At least not without pressure.

An easy way to greatly reduce bacteria on a spore print is to simply wait to use it. Most bacteria will die from desiccation but the mushroom spores will remain viable.

Why not follow the methodology of that paper and just dunk the spores in 3% peroxide for 10s, then dilute with sterile water by a large factor until the peroxide concentration is very low before you streak onto agar? I'm not totally sure how low that would be, given that when you add peroxide to agar the recommended concentration is something like 0.02%, but anyway it seems easier than trying to wash the spores by pumping them through a mesh.

The extra weight of water on top creates pressure but at some point before it completely empties it will stop flowing and you'll have to wait for the rest to evaporate. To show you what I mean here's a pic of my inoculation loop. The pore is far greater than 5 microns.


I think the idea of cleaning the spores with something that's toxic to them is probably a bad idea. Alcohol will kill bacteria but have little effect on spores it'd probably be better choice of disinfectant. If I were to try cleaning the spores I would just place a couple drops on the spores at a very low concentration in a sterile environment like a petri dish and wait for the water and peroxide to evaporate on their own.
