Hi Forum,

A couple of days ago I inoculated some petri dishes with Lion's Mane (10 of them), Garden Giant (5 of them), and Shiitake (5 of them too.)

Today I see growth in all 10 LM dishes, 3 of the GG's but no growth in any of the Shiitake dishes.

I have had them in the dark though so I have lit a lamp from now on.

What do you think that the lack of growth in the Shiitake dishes can be due to?

If I continue to see no growth in a petri dish, can I wipe the surface clean and inoculate once more with the same of another culture? They were pretty wet, a lot of condensation, can this be an inhibiting factor? All inoculations were with liquid culture.

Shiitake can be slow. When did you inoculate? I wouldn't even think of trying to wipe down the agar surface to reuse it. If it was a no-pour agar dish, you could just PC is again and reinoculate. But if its a disposable petri, give it more time, and if it winds up being contaminated, throw it away.

You said there was a lot of water\condensation. Is there pooling water on the agar surface or just on the top petri dish cover? If its just condensation on the cover you can put the petri upside down on a hotplate(on very low heat) to clear up the condensation after a while.

Actually now it was a full week ago. Maybe I should pressure cook again, is it enough to do only 10 minutes of 15 PSI? I do not want to caramelize the sugars in the agar. (They are glass petris so I was thinking I'll just stick it inside the PC wrapped in foil.)

I'll see what I can do about the water inside the dishes, there is some water floating around inside the dishes, some drops, not pools.

The ones where it has gotten hold, do I turn them agar side up? (I just remembered that this is one recommendation that I forgot, is it vitally important to do that?)

i have seen people get away with glass petri no pour, i think it seems crazy!!  but if it works, then more power to ya!  i would toss the ingredients, and do a new batch.

i use more agar in my recipes now so its stiffer, easier to do transfers, and also helps if any condensation occurs, or it seems to anyway, if anything drips onto the agar, pooling is less likely with stiffer agar from what i can tell.  i use 11.5g per 500ml now instead of 10g agar.

i would expect to see shiitaki LC at least start to show growth within a few days.  if u don't see anything on the plate still, i would contact the vendor and ask for a replacement.  i have gotten replacements from vendors here for edible liquid cultures that did not arrive viable.  they are pretty understanding.
