Bacteria Damage Control?
    #2120626 - 11/18/03 05:24 PM (20 years, 4 months ago)

I recently did an agar to agar transfer of my shitake myclium that was showing strong rhiz growth. Unforchanatly on 3 out of 6 dishes water dropplets formed (damn bacteria) on the agar triangle placed in the middle (I put the mycilium side down it handn't grown compleatly through). Since faste rhiz. growth has occured around the transfered piece and none of the area shows any sign of contaimination.

My question is should I bother with the growth out from the transfered wedge and put that in a substrate jar? Can I do any damage control? Any tips would be appriciated!

Thanks,
Saffron

Keep moving clean zones to fresh plates, untill
you have left the bacteria behind, are you transphereing to
agar containing h2o2, or you could research antibiotics that
can be added to your agar soloution.

You shouldnt use bacterialy contaminated agar in jars though.

I would transfer to a plate with antibiotics added to the agar and let them fight it out. I have seperated clean mycelium from contamination, but it took a lot of time and effort, plus I had access to good lab equipment at the time. The biggest problem I had was figuring out how to change the environment so that the mycelium would grow faster than the contamination. Once you do have clean growth, you will want to do a few more transfers to ensure that nothing is hidding.
