I've now got two monotub runs under my belt and I'm interested in getting more serious about cultivation. Right now I have six, almost finished, grain jars that were prepared using a multispore liquid culture. I also have 2 PF Tek jars that are about halfway done, inoculated with the same liquid culture. I want to move past using MS spawn and try and isolate a strain from what I've got. My current plan is to use the 6 jars to spawn two 27qt monos, then use spores from the best looking fruits to try to isolate on agar. The PF jars will be used to practice making slurry and spawn bags while I wait. This sounds like a good plan to practice some more advanced cultivation methods, right?

And since I hadn't seen any contams in my jars since I started, I was really sloppy with a BRF jar just to see.

Take a few colonized grains from your healthier faster jars and put them on agar. This will give you some sectors to work with quite fast, maybe even right off the bat. This will save you from doing a dozen transfers or so just to see sectoring. Way less time consuming and way fewer plates needed

Right on, that makes sense. Gets me going on agar sooner rather than later. Is there any difference between isolating from some colonized spawn and isolating from an actual fruit? Results-wise, that is, not procedure-wise.

From a fruit is cloning. Is less stable but more predictable. True isolates are going to be unpredictable. Most will perform terrible. But around one in ten will be stellar. So carefully labeling and testing each one will be important.

From what you say it sounds like cloning is what I was thinking of. What do you mean when you say "unpredictable" and "unstable"..is there a good place to start reading to develop a base of knowledge on mycology? I feel like I'm missing a lot of basic information about mushrooms/fungi in general.

Clones are essentially older cells that have already expanded a fair amount. So they are more prone to senscence. They are also comprised of many sets of genetics which may inadvertently be stripped out. This may result in better or poorer performance. They are a good indicator of fruiting genetics tho so yiu can usually expect certain results based on the clone you took.
With an isolate you won't have any idea what you have until you fruit. It could be amazing, it could be shit.
You can learn all of this on the site. Use the search engine. Its all here. Or you could buy some books like The Mushroom Cultivator. Be aware that books often carry outdated info so double check the quality of what you read elsewhere before taking it as gospel.
