Do you have a vid for taking spore prints, then adding to a solution? I reccon you should journal this with an index, its just the best thing on the web.

You can ascertain the steps from here, just don't iso your needle or your cup. Wrap your shot glass in foil and PC it. I bring my PC to my SAB and then open it real quick and load everything into my clean SAB via the arm holes. Then I get the print, scrape after flame sterilizing my scraper and get them in the shot glass (After taking off the foil being really careful not to put my finger inside).
Then shoot the water, suck, shoot, suck, shoot, etc.
Munch you should update this though.

Sterile tap water that I PCed. Don't use more than a pint of water per plate or it can get too diluted

I just watched the agar to agar transfer. Shouldn't you cool the blade in the receiving end since it's much more likely to be clean? The plate with the growth could have something bad on it.

IDK. I'm sure it is debatable, though. One counter argument to that might be that you are moving around a cooled blade in a moderately clean environment more by going back and forth between the plates. I have honestly never had any problems doing this, though. Contaminates on my transfer plates are very rare.

When I make a transfer. I don't even bother cooling the blade. I just start cutting out my wedge while the blade is still hot. You singe the first bit of mycelium but you leave enough healthy mycelium to start growth on the new plate.

Probably. I have concerns about everything, heh. I've just always read/heard that to avoid contams, you'd dip into the receiving plate since it's cleaner. I think RR was an advocate of that. Either way, I'm sure your contam rate is pretty low so I doubt it's that much of a risk, if any. Awesome vids, I like them a lot.

The way you do it seems fine to me too. If there were conams on the portion of the plate where you are cooling your blade you would see them growing. That's sort of the point of agar.

I didn't cool the blade for that transfer, everything worked out just fine. the second picture is the same thing zoomed in
