A nail, hard drive magnet, and a computer fan is all you need to set your self up with a stirrer. LC growth is sooo much faster when it is stirred frequently. Shaking instead of stirring isn't the best idea as well a nail has its edges which allow it to chop up the mycelium towards the finish of the lc when it gets super thick and even then you might face problems getting the mycelium into a syringe...

What do define as frequently stirring? One time a day of more often?

Noc'd up two jars of oats with that LC the other day..two days and there's already some significant growth even though it got cold yesterday - I'm already impressed with using LC this is my first time I always just use agar wedges to inoculate
If there is explosive growth within the next couple days I'll throw up pics

This is what became of the WBS that the WBS GWLC was made out of.

Loaded the gallon sized jars with the half gallon jars, and a few quart jars into the AA 941, and pressure sterilized the media for 2 hours at 20 psi.

The stirplate came in yesterday in the middle of lab.  Dropped lab for a few hours to play with the new stirplate, and figure out how it works.

While figuring out the stirplate, the decision was made to break out the frozen WBS Grain Water, and make a few LC's out of the concoction.

Since the GW was a frozen block of hardened ice, and some was needed right away, a sauce pan was filled half way with water, and heated with the frozen grain water inside of the sauce pan as to instantly thaw the frozen grain water.  Small mouth jar lid rings were used in the bottom of the sauce pan as the grain water was heated so that the bottom of the media storage vessel didn't make direct contact with the super-heated floor of the sauce pan.

The GW was poured into it's respected media vessel as soon as it became thawed.  The vessels were filled half way full with the 50% diluted GW concoction, then filled the rest of the way up with water for a dilution ratio of 1:4.  The vessels were then capped with the closures fitted with the donated Whatman filters.

placed into the sterilizer, and steam pressurized for 50 minutes at 15-17 psi.

Got some Pan Stip, and Blue Oyster mushroom live liquid culture going on.  The Blue Oyster mycelium has almost fully colonized the media, and the Pan Stip myce is colonizing right along.

Pan Stip Liquid Culture.

Blue Oyster Mycleium Liquid Culture.

The Blue Oyster mycelium is scheduled to be aspired into syringes today, or tomorrow, and be shipped to Toronto to be donated to a How to Grow Blue Oyster Mushrooms class there ASSUMING everything is all good with customs.  Going to the PO Monday to check what customs has to say about the sending of live, liquid culture syringes to Canada.

The undiluted Red Milo media has FINALLY got observable growth.

Can't really see it in the picture, but the growth goes up to the first line, and is SLOWLY beginning to clear up the media.  Everything looks happy, and a happy vibe from the undiluted Red Milo is had.

The original undiluted rye GWLC is 7 days into spawn run (colonization of the bulk substrate) colonizing away.  More on the bulk substrate spawning later.

Again, thanks for checking this project out, and as usual, more to come!

Looks like the cat's outta the bag now, great stuff loaf

My wheat LC has sat for three days unobserved but I checked it this morning before I left for work and I already see the wedge fuzzing up, won't be long now. I will toss up a pic here when I get back. Its not as rich as yours but at 2 parts water 1 part gw its still plenty rich.

Here goes a look at 2 of the 6 minimonotubs generated using the original rye end of the boil-cycle grain soak water.

They are at day 8 of colonization, and ALL of them are displaying hyphal knots/"knotting-up".

It'll be just a few more days before the these minimonotubs are introduced into fruiting conditions.

Here's a look at the monoculture intended for the inoculation of 2 master culture slants, 1 quart sized grain jar, and more importantly, this monoculture will be exclusively inoculating the freshly sterilized liquid media using WBS end of the boil-cycle grain soak water with the addition magnetic stirbars so the culture can be stirred using a magnetic stirplate INSTEAD of the hands.

Have a 3 more monocultures, but this one looks the best, in the opinion of the individual cultivator.  Next stop, FRUITS!  Thanks again for checking this out.  You know the rest.

More to come!

Whatman filters ftw. Do you use rubber gasket for those plastic lids? What will you be inoculating these with?

What these mediums will be inoculated with has been answered in the post above.

Rubber gaskets for the lids are NOT used, nor are they necessary for the needs of this researcher.  Dabbled with them in the beginning.  They would be used if the cultures were shaken more than once before aspiration.  Liquid cultures in this research lab are only shaken once, by hand, JUST before aspiration, and in front of a flowhood.  Shaking the culture in front of the flowhood allows for media to safely leak through the closure seal, and get the filter wet.  Once the culture is removed from the laminar flow of the flowhood though, once the shake has been administered, the culture is considered contaminated.  The culture is then discarded.

Not worried about air being drawn into the medium during the return to atmospheric pressure by the sterilization vessel either.  For the air being drawn in will take the path of least resistance.  That's through the media vessel's filtration system.

About to change the fact that liquid cultures are only shaken once, AND the fact that they are shaken by hand with the addition of the magnetic stirplate, and magnetic stirbar, as well.  Will keep you posted.

So my tamp LC turned out GREAT!
I've decided to use up more of it and knock up atleast 4 more jars
Here's my question and or something I'm about to do... I'm thinking of squirting a cc or so of the LC  into a brand new LC to grow and preserve it further. Bad idea?

I wouldn't,  you would be introducing vectors that are unnecessary. If you want to save the culture then inoculate a plate then transfer from the plate to a fresh LC and a master slant to store long term.

I'd like to see a picture of this settup I have a few old computer fans around and am sure I can scare up a magnet gotta see I may even have an ols hardrive lying around.

There's a bunch of videos on YouTube that go into building one for yourself. Brewers make them for yeast starters. Personally I like the one made out of a wooden cigar box.I just need to find a hard drive, or a couple good rare earth magnets.
