    #2116554 - 11/17/03 12:54 PM (20 years, 4 months ago)

Ok...so lately i have been noticing that my jars are not colonizing very fast...so i am wondering about doing a grain 2 grain transfer..or getting into agar...does anybody have anytips for me on agar...i was thinking about buying one of those agar sets from micron..but im just wondering what peoples experiences are with it..thanks

ive been doing the pf method for my jars but i am looking to expand my credentials...

Why don't you just make really potent inoculum and inoculate your jars with that? Also, minimizing the water content can speed things up and then you can just case that (you want more water if you're growing PF style.)

give me a clue on some really potent innoculum,..or do you mean...make my own syuring and make the mix really purple..by adding more spores to the syringe...or doing a liquid culture...with mycelium in the syringe....i tried the karo tek..and had no success...but it might be because i used tap water instead of h20...

I think he means tap water as opposed to distilled water.

what would be wrong with clear, clean and untreated water ? I mean it should not be a problem after being PC'd, or do I miss something ?

well..i think that distilled water is a thousand times cleaner then regular old tap water...i havent had too many problems with contams..but i do wonder if it is from the water...

if you PC'd it properly there can't hardly be any contamination left. There still will be some minerals in there, but that would be the only difference to distilled water that I can think of.

I'm guessing why the karo tek failed for you is the fact that liquid cultures contaminate extremly easily. Judging from your low number of posts, I'm guessing you don't have a ton of experience. Don't get me wrong, I'm not too much further ahead of ya, but my point remains; inexperienced growers attempting to do a very difficult tek (difficult in the idea of keeping sterile) will most likely fail. I failed when I tried the honey tek. Newbies tend to use shortcuts like I used to do, until I realized that the shortcuts only make you go right back to the drawing board when you shit gets contaminated. So now I do everything right, built a glovebox, use enough lysol to kill anything that dares get near my shit, and boil things to shit (being without a PC, fractional steralization is a nifty, yet painstakingly long process).

So my advice, try the karo tek again, but learn from your mistakes. Use distilled water, build a glove box, get some tyvek from the post office, and steralize that shit to high hell. Good luck.

ragadinks the problem with tap water is probably clorine treatment or maybe ph, if you dont test it. Comparing the sterility of distilled and tap water is pointless as both must be 'claved or PCed anyway.

yes, PH is a point I did not think of.
Is there a recommended optimum PH for growing mycelium ?

When I mentioned sterility, I didn't mean that of the water, I meant of the entire grow in general.

***EDIT***

My bad, I didnt see that the comment of sterilizing water you made was in reference to something said way before my comment.  :smirk:
