Well... generally if the plate itself was dirty then you would have "satellite colonies" but you don't have that here...if that is mold in the center of the plates then it was present when the transfers were made...not many molds have rhizomorphic mycelium so that is probably your best bet for cleaning up the culture via transfer

that is one of many indicators of a dirty plate. but not the most important. the easiest contams to see and get rid of are satellite colonies. but hidden contams are the bane of my existance

mold sucks when it get's wrapped up in mycelium. check out these fucked up plates. i went back to new germ plates after seeing mold win the war.

Damn. I can see in JHOVA pictures the almost clear mycelium that is around the edges of the white as well, all my plates have that in a certain light. Maybe the APE swap had some nasties on it from the get go
Maybe I'll just transfer once more, and see if these plates pin or even keep going

The point of agar work is to clean up dirty culture...there should never be a point when you are stuck ... invisible contaminations on agar? I mean it will sporulate eventually no matter what. I know a lot of you guys aren't using flowhoods but a dirty agar plate is never a show stopper...unless you are just outright bad at sterile technique or you just don't understand the fundamentals (the "why's?") of it all

Are thes contams in the cultures? I have 3 different cultures and they keep transferring like this. I actually finally took a plate to grains and ithose grains are in coir right now.
I swear no matter how I transfer it, the cultures look like this. Maybe I need to go to a SAB instead of a hood?

That just looks like sectoring to me. Is this supposed to be an isolate? If not keep in mind as the individual strains seperate the mycelium will look different from strain to strain...we are generally disposed to going for the most rhizomorphic strains as far as cubes go.I dont think that is contamination.it can take many transfers to get down to a monoculture

where would you take a wedge of the agar i posted myco217? Those are T4’s. Its less work in my opinion to streak a fresh plate and start wth less mess.
If you have some pics post em of bad contams you have gotten away from.

I would transfer from the outermost edge at the 9 o'clock position on your second plate pic
I transferred away from this and cleaned it up:

That being the case...contamination shouldn't be a constant hurdle...it should be something you overcome through learning proper sterile technique which myself and several AMU members have links to instructional videos in their sigs

I try to use the first clean plate, keeping initial MS tubs as genetically variant as possible personally Clone several from that tub for side by side's in shoeboxes, picking and slanting my favorite for further isolation projects, based on colonization speed, first flush yeild, and potency... Here's what I've got going on... 5 tubs in a GH. One had the colonized WBS fortified with Brf, the rest all the same otherwise.
...and apparently one is growing upside down.
