Yeah, that's weird for sure.  Keep an eye on thing.  Report on how the media develops, please. 

One thing that is noted right off hand is the handling of a lot of peoples liquid mediums with their bare hands.  Handing your media vessels with out alcohol soaked, latex gloves on is a HUGE vector for contamination, and what's the most contamination sensitive media in this hobby today?

I usually glove up myself to pick up my LC jars as well. I also parafilm the lids edge. I might be paranoid but :shrug:

SAME.  All media jars are ONLY handled with alcohol soaked, latex covered hands.

No.  The hands, next to the feet are FULL of ALL KINDS of bacteria.  Stick your bare finger on one of your freshly sterilized agar mediums and see what happens.  Better safe than sorry.

The feet are actually an exit point for our bodies to excrete internal contaminates.  There are foot treatments you can get that ACTUALLY encourage the aforementioned process.

Would not suggest inoculating anything with that contaminated liquid culture, unless you are trying to observe mold growth.  Otherwise, you are just going to spread the contamination through out your home once it sporeulates.

A look at the diluted Red Milo GWLC just before the "shake and aspiration" procedure. 



This media is PACKED full of liquid myclial culture.  Probably over 100cc's.  This media generated a MUCH thicker mycelial culture than the non diluted Rye Boil Water.  My guess as to the reason for the denser, faster colonization was due to the lesser amount of nutrition in the media.

Generating the liquid mycelium in a nutrient thick solution though creates a HUNGRY mycelium that once it hits the grain medium with a far less concentration of nutrition than the young culture is used to having available and consuming, the mycelium agressively colonizes to satisfy the mycelium's INSATIABLE hunger.  An added bonus with the GWLC is that you can get the culture used to the specific food source, or grain the liquid culture intends to be inoculated to.  That's a double threat.

Drew 60cc's of the culture into a 60cc syringe for short-term storage purposes, and discarded the rest of the culture.

 

It's like the Rye GWLC began to recover AS SOON as it left the syringe before it even hit the grains, so I am anxious to see how hard this Red Milo grain water liquid mycelium PUNISHES the grains its put to. 

In relating news, the original experiment with the non diluted rye GWLC has been spawned to bulk substrate materials comprised of a 50/50 mix of pasteurized horse manure, and lizard substrate coir prepared via Damion 50/50's Elementary Coir Tek inside of 27 quart, Sterilite clear plastic storage totes with clear, latching lid modified into minimono tub style fruiting chambers, and are three days out from inoculation INCUBATING at a fluctuating temperature range of 68-72F. 



WHAT HAPPENED TO ALL YOU OTHER MEMBERS' MAD GWLC EXPERIMENTS!??  It's ok if they contaminated.  Contaminated liquid cultures should be reported, and documented with pictures too so that a contamination guide to visually contaminated LC's can be started, and referenced until completed.  Reporting on the failures also spawns ideas on how to solve the problems that created the vector for the contamination to the media and culture.

Forgot to update. The first set I did was too thick but as I poured it out I saw mycelium so we were good to go, I juts couldn't see anything. With that being said I did Grain water agar a couple days ago and now have the hang of oats.

Will be retrying this soon.

The red milo looks choice :awesomenod: I am still finishing up with a few malt LC's and decided I am done with the SITR (dissappointing to say the least) so my next batch will wheat. Got some frozen and waiting :thumbup:

Yea, I was going to laugh at you earlier when you first reported experimenting with sugar in the raw liquid culture, but kept my mouth shut instead.  :lol:

Malt extract is the favorite for making liquid media out of.  Just inoculated a few grain bags with some clone PESH LC in some Malt extract media broth.  Have several hundred CC's of some Blue Oyster LC that is being donated to a Blue Oyster class in Toronto.  Some will be given away in the Marketplace.

Just about ready to replace the malt extract as the liquid culturing media of choice though with this GWLC phenomenon.  There are three major advantages that I can see so far of using the grain boil water as the liquid culturing media of choice over Malt extract.

Shit man I gotta try everything once. I tried honey once and it sucked. Tried Karo and it sucked. So I figured I had little to loose. SITR does look purty tho :tongue:

Like violet I started to use GW for agar way back when looking for alternatives to MEA and PDA. I noticed how good it performed so I thought what the hell, might as well make LC outta it.

Yeah, I stared at the grain water a bunch of times before we had that conversation, then decided to run it.  IME, all those Karo, Sitr, and honey ect LC's suck. 

I disagree the aesthetic pleasure you see in SITR.  Could be an acquired taste, but I like my LC's golden.  Starting to acquire a taste for LC's being a little darker now.  :hehehe:

Violet, What's this "grain agar" you speak of, do you have a recipe??  That sounds AWESOME!
