The Beautiful Beginning.
    #21135735 - 01/17/15 09:20 PM (10 years, 1 day ago)

Hello, Shroomery! This is the beginning of my first cultivation operation. I have been reading threads from this forum for the past year in preparation for this wonderful experiment. I am following the Pf Tek, as per recommendation to accommodate me as a noob. I mixed roughly six cups of vermiculite to three cups home-ground BRF with three cups purified bottled water. I then sterilized the jars in two seperate sterilizations due to small pots. I constructed a still air box using cardboard, cling wrap, and trash bags as the impromptu gloves. I sanitized my gloved hands with 70% isopropyl alcohol. Six half-pint jars received Pf Original (Amazonians? I have heard controversy as to the actual strain that this is. Anybody have any information on that?), and six half-pint jars received PC B+ spores. This is how far I have currently gotten. I am a little worried that my substrate may be too dry, however I have been told otherwise. It has been about 24 hours, novisible changes, but i dont expect there will be any visible rhizomorphic growth within the next three or four days. Any comments or criticisms are appreciated. Thanks for the read!

Click [pics > upload > choose files] to upload pics

Then, after your upload finishes, just click on the link under the photo and it will copy and you can paste it to your message.

It can take a week or two before you see any noticeable growth. Ambient light in a bedroom has been proven to be beneficial to mycelium, so just keep your jars on a shelf or somewhere they can get indirect light.

If your BRF+verm was at field capacity before sterilization, your moisture content should be fine  And as long as it can finish colonizing, you will be good, because you should dunk & roll before fruiting. The "dunk" will re-hydrate the mycelium.

As for the strain question, from my knowledge, PF classic is Amazonian and it's also sold under another name - a city in mexico - the actual name escapes me right now. Grain of salt; I could be wrong..

Good luck with your grow, my friend!

I've never heard that PF classic and Amazon are the same.  Can anybody provide a little background on this?  I have some PF classic almost ready to fruit and haven't really checked it out at all, but have been generally interested in eventually doing an amazon grow.

It's all kind of in the air, from what I know. I believe it was Roadkill back in the day making a fuss saying pf and amazonian are the same. PF said their were different but thought people started reselling PF under the name amazonian.. Blah blah. Search it up if you're really interested. The other one is matias romero I believe, which people say is the second alias.

What I think it is, is that people started reselling PF under the name amazonian like pf said, then it all became confused and fucked and some vendors kept the name amazonian while other called it PF classic and then some assholes just threw the name matias romero in there for no fucking reason. IDK though, I'm just talking out of my blotched, shitty, half hallucinated memory of posts I read a long time ago. You'd be better off searching around, specifically with roadkill's posts on the subject

in my experience, too dry is better than too wet, but when following the tek you shouldn't have any problems.

One glove was stationary, while one was one my hand. I used the detached glove hole to insert the syringes after sterilizations and between inoculations. I sterilized the needle with a Bic lighter once between each jar as to prevent any contamination. Does that sound like the appropriate procedure?

Yea this. Attached gloves stir up air currents

But for PF tek you'll be fine this time, its one if the few things you could do in open air if you wanted. Of course. Its always best to use a still air box.

The first grow is always most exciting and it looks like you did every thing right, good luck!

Looks good, man, keep it up. First getting your feet wet in this hobby is awesome. My advice to you would be to try everything that interests you. There's a lot of stuff out there that's good and a lot that's bad, but firsthand experience trumps any anecdote over the internet. Agar work is super fun and really easy inside a proper SAB, probably one of the more rewarding parts of this hobby imo.

If you need a way to kill some time you could get your hands on RR's videos and watch them, they're bursting with information. Get your hands on some mushroom literature and learn some stuff.

UPDATE: Four or five of my jars appear to be colonizing swimmingly, around 25-50 percent surface colonization has been achieved for these guys. However, the rest of my jars are closer to around 5-10 percent colonization. I believe there is only one that has yet to exhibit signs of life thus far. My brother is taking care of half of the babies at his house, and I feel like I am a parent who has been subject to a joint-custody sort of deal. I am very excited for these next two or three weeks of colonization, but moreover for fruiting to follow. THIS IS AN INCREDIBLE EXPERIENCE.

So, I was checking my babies out this morning, and i am really confused by their diversity of growth. One of my jars is around 35ish percent surface-colonization, one is around 10 percent, and two have only dots of mycelium showing. So, my query is this: what is making this one jar in particular grow so much faster than its counterparts? I will upload pictures of my four jars this evening for further reckoning. I appreciate it!

It is indeed a splendor to behold. All the way up until I actually saw my pins develope into a cap and stem. I did not believe it was a reality it was years ago but I still remember saying to myself this can't b possible its all a scam the prints the syringes the pictures there's no way this is possible for a normal dude. Only scientists could make this a reality. I didn't even think my box of goodies would arrive in the discreet little homestead book company box. Then it did. Inside was a print agar presterilized petris (20) count inoc loop scalpel bag o composted peat and guide book. Even after my tranfers and grain nocs and myc groth I still said this isn't going happen and then even after my casing layer colinised and primordia formed I was skeptical even as I watched the small white dots form and swell there was disbelief but once I could discern my frst stem and cap I felt like a god and that feeling my friend never ends every pin thereafter is one magical moment after another.  Welcome to the counterculture

Thanks for the welcoming, Proffessor!
You really seem awesome; I like your style. However, I am having a problem. One of my jars reached 100% surface colonization yesterday, but there is a slightly off putting scent coming from the jar. Its something akin to sour laundry maybe? The smell is not strong really, but it is there. There is also a slight brown spot on the mycelium. This jar grew from 10% to 100% over three days, so I suspect it coukd just be mycelial waste? If it was a bacterial contaminant, the jar would not have colonized right? Picture coming in moments. I appreciate it guys.

Well a good thumb rule is. If there is any odor other than fresh smelling myc then there's an issue. I now live by a strict code of if there is any doubt as to its viability or purity then there is no doubt. Becuz  fresh noncontamd cultures smell is unmistakable. And it seems everytime except for a very few that I went foward with a questionable spawn jar or brf cake it just contamd and was a wasted time energy and space.that jar looks well colonised but a lot of waste water in the jar and with such a speedy colonise time if ur havg issues u mite lower incub temps a bit

The incubation temperature is only 70-75 degrees Fahrenheit. However, I honestly dont know what "Fresh mycelial" smell is. The aroma isn't what I would consider sour, or sweet, it's just not the smell of the original brf cake prior to inoculation. Should the smell be closer to a musty smell? I am going to keep it in incubation for sure, and see if it forms any invitro pins. Is this good? At what point would it become apparent that the jar is contaminated? Tomorrow will mark three weeks of colonization, and I will be distraught if the consensus is to trash my baby. Second opinions? Further opinions? Thanks for the input professor, and it's not your fault that your information isn't what I wanted to read. I really do appreciate it, man. Im just bummed out.

Hello, again, everybody! I am here with a handful of good news, and a handful of bad. First, the bad: two of my four jars are likely contaminated. I know, certainly, that one is because of it's smell. The other, however, is different. Initially, it's growth looked stalled with three spots of fuzzy white mycellium. Now however, those three spots appear to be dead. They are not black, but kind of the color of the original substrate. The rest of the jar is now colonized, but the original sites of colonization seem dead. On the lighter side: I spotted my first primordia ten minutes ago! I am now preparing to dunk my two fair cakes, and prepare them for the fruiting chamber. Pics are incoming. One question: Is it necessary for my cakes to be chilled during the dunk, or will ambient room temperature be acceptable?

We are almost there, boys! This is incredible.

Got pics? They would help a lot. And room temp water is fine.

Certainly, good sir!
This is a picture of the jar which is being questioned. The question is not if something is wrong, (Because something obviously isn't right), but is it still worth birthing and fruiting? Anyways:

This is a shot of my first primordia EVER!

Here is one without the jar, prior to the dunk:

It even looks like he has a little brother! Let me know what you guys think.

Also, the black on the outside of the jar is where I traced the mycelial growth with a sharpie during their third week in order to assure myself they had not stalled. It's nothing malicious.

Since you have other healthy cakes there, I would toss the contamed one. You'll have plenty of mushrooms soon enough, no worries.

I figured as much... Do you have any idea what caused this? Perhaps a bacterial infection? It is worth noting that the two to contaminate were my pf original, and my B+ went along swimmingly. Would a shroomery sponsor refund me for a contaminated ms syringe? Thank for the congratulations, brother!

Probably improper sterile technique not the syringe. It's possible it is though, that's why we put spores to agar to separate the contams from the healthy myc and ensure health.

Yeah, you're probably right. It is alright, though. I plan to crumble my two contaminated cakes beneath my rabbit hutch, outside. That is, in my opinion, a better fate than a garbage can, even if it only lets them decompose.

I was skimming and then hit the back button to get back to my threads pages but the words "I plan to crumble my two contamined cakes" clicked.

You should have seen how fast I hit the forward button .

Good plan.

If the weather is good and you mist a little every now and then yes.

Cubes are a beast in the wild. They're only so susceptible to contams in our home cultivation because our methods create a perfect breeding ground for not only mycelium but molds as well. We don't mimick nature, we try to improve on it.

So outside just make sure the area is a little wet every now and then and not too dry and they'll grow.
