Help! Agar wedge grain inoculation- A sticky situation

After I drop an agar wedge into a grain jar sometimes it sticks to

the upper side of the jar. This does not happen all the time. I try to

carefully shake the jar so the wedge gets covered with grain (ideally

ending up in the middle or bottom) but sometimes it sticks to the

side, when that happens I just turn the jar on its side to colonize so

that the wedge that is stuck to the jar is at least covered with

grain. It's not really that big of a problem but it is a problem that

I would like to solve if I could. The reason I want the wedge

completely covered is because the leap of is quicker and the jar

colonizes quicker than if I just dropped the wedge in the jar. Does

anyone else have this issue, or even better, does anyone know how to

solve it? I was thinking about adding gypsum to the jar but I don't

know if that will help.

Just cut your agar into smaller pieces, so there's more inoculation points. Then you won't worry, if one piece is stuck to the jar.

Thanks for the quick reply. That's not a bad idea, and I will try it. The only problem I see with that plan is that it seems that it would create more work and more of a window for contamination. Let's say I'm inoculating 32 jars with four petri dishes. I typically cut my plate into 8 wedges like a pizza and each wedge inoculates 1 jar. That means there are 32 transfers I have to make, if I cut each wedge into smaller pieces the number of transfers skyrockets (a lot more work) as does the chances of contamination.

what ayahuasquero said. i started cutting that size chunk into 5 or 6 smaller pieces to minimize this issue. i slice up the petri all at once, then for each jar i stack 4 or 5 wedges on my scalpel like a shishkabob. i also started flame sterilizing my scalpel between every jar when i switched to this method because sometimes i have to use the side of the jar to get all the little agar pieces off. better safe than sorry. i still get pieces stuck to the lid and sides sometimes but its not a big deal if its one or two out of five. if someone else has a better idea, i'm all ears

32 jars sounds like a PIA.  I have had great success with agar -> master grain jars -> grain XLS filter patch bags (lay them on their side).  10-12 quarts each bag that is very fast.  Just got sick of so many jars...

I usually use one wedge per master jar that is about the size of a dime and they run quickly and don't get stuck.  I can use a single dish many more times doing this.

I don't actually do 32 jars. So, are you saying that because of the smaller size of the agar it does not stick? Or are you saying you don't shake em?

It does not stick.  I shake them after dropping it to get the wedge in the grain.  Shake again at about 10%

I also cut my agar into much smaller pieces; usually i use about one square centimeter per jar cut into four pieces and scooped all at once with a scalpel.

If I had 32 jars to inoculated I'd be doing g2g inoculations.

i don't shake anymore when i put wedges in.  i wait around 3 days, let it fuzz up, then shake like hell.  it was getting stuck on the glass, even when i poured pretty thick.
i like going agar>1pint>3 quart jars>6 nine pound bags.
the pint jars go QUICK!
