slants/vacutainers/storage

hey guys,
so sometimes when i receive cultures from northwest and others, the slant will be completely screwed on tight, and will also have parafilm. i was under the impression that even after the culture has colonized the slant, it should still remain slightly unscrewed, even during cold storage, for GE. is this wrong?
and as far as vacutainers, or other "rubber stopper" style slants, are those just pushed all the way down prior to placement in the fridge?

i would screw them tight then unscrew 1/4 turn, then parafilm. but that's just me. hardly any of my cultures are in slants, thanks for reminding me to do it!

yea thats what i normally do, 1/4 back from tight. i need new slants, i have those small ones, like the aloha tubes they send cultures in. i got a nice metal tool that is long, with a spoon on the end, but its almost too big to get in there! also got these little tubes i think maybe they hold 5ml water. was wondering what i could use them for, maybe distilled water storage?

yeah i guess put wedges in there. im going to make some water agar dishes and chop them up for storage like you're talking about before making proper slants i guess. especially for these tropicals, they've been sitting around at room temp for months!

how do u use water agar? is that what u would need for distilled water storage?

Transfer a wedge to water agar (same as any agar recipe but leave the nutrients out) and let it run. Take a wedge from the water agar and transfer to your sterilized distilled water tubes. Seal them tight and leave at room temp.

Can you refrigerate or does it not matter? Not with warm species obviously

It isn't needed but shouldn't hurt anything. I am always fighting for room in the fridge so they don't go in there.
