Storing jars after sterilization and a modified still air box colonizing chamber?
    #21015349 - 12/23/14 02:25 AM (9 years, 4 months ago)

Two parts for an idea I had.

First part is how can I better store my 1/2pint jars after I clean them after use? I've run into contams my first two attempts. I've had some ideas to better protect my next try from contams. I'm going to throw my jars in the dish washer on heavy wash with sterilization setting turned on after wash  because there is still some hard mycelium or mold on them. I'll wipe them down with alcohol wipes than once dry I'm going to sterilize them in the oven. I'm going to wash the lids and metal rings in a bath of ISO Alcohol than once the jars are done sterilizing and the lids/metal rings are done in the bath I'm going to put them back on the jars than wrap every jar with plastic wrap to keep them safe before putting my PF cake mix in them and steam sterilizing.

Part Two:
I was thinking of using a still air box this time for inoculation. I ruined my last two attempts by inoculating in open air. I somehow missed the recommendation of wiping the needle with an alcohol pad between jars. I'm going to wipe the needle with an alcohol pad between each of the 4 holes per jar, and flame sterilize between each jar. For the colonizing stage I want to use my still air box to keep the jars in. I want to know if I can use the lid of the rubbermaid container for this. By cutting a few holes in the lid that I wont be using for the still air box during inoculation I will place coffee filters over the holes and tape them in place with pore tape. While I will still have micro pore tape over the holes in the lids of the jars to prevent contams from getting inside (even though the verm layer should help with this) I still want to better protect any outside contams from my house getting inside the still air box. Will coffee filters work for this purpose?

If there is anything else I'm missing that might be a source of contaminants please let me know. I'm also going to sterilize the vermiculite in the oven before preparing my cakes. I'll also be using purified bottled water and I hope its not junk municipality water bottled up and perpetrated to be purified. Could I boil the water before using it to mix up my cakes so I know the water itself is not contaminated?

I've failed twice already. My poor technique has to be the cause. I think me using the still air box right side up with the lid attached with holes with coffee filters might be a decent idea for more protection instead of just having them sit on a shelf. Third times a charm I hope I don't see contaminants a few days after inoculation. I obsessed over the air temp the last two tries so I'm discarding my thermometers to quit doing this. I've read someone claim they gave colonizing jars to a friend who put them in a garage that was 30 degrees fehrenit at night and they still colonized. My house cant possible get that cold.

This is all unnecessary, proper sterile technique is all you need.

I'm not gonna list this stuff in order but here:

There's no point in cooking verm before you mix it in your substrate, for one you're already sterilizing your jars, you can't have anything double sterile. Even if you sterilized your verm, as soon as it is exposed to open air it's dirty again.

Tap water is perfectly fine, no point in bottled/boiled water.

Unless you're just getting substrate off of your needle, never wipe with alcohol, and if you do, still flame sterilize afterwards. Flame sterilize between every hole, and don't wipe it with anything afterwards, if you do, then the needle that you now just made sterile, will be dirty again. Alcohol sanitizes, not sterilizes.

Mix up your substrate, place in your jars, use your dry verm as a filter, steam sterilize/pc them, inoculate in a still air box, flaming your needle outside of your sab between every hole, and every jar, then take them out, place them on a shelf, and wait.

Just follow mushroomvideos.com part 1-4 exactly, the extra steps you're trying to add will just waste your time, and make you more likely to mess up.

If I missed anything you're curious about, go ahead and post.
Good luck.

:whathesaid:

That too, just pc/steam sterilize your bad jars as normal, then wash them out with soap and water like your washing any normal drinking glass .
