So sorry taGyo, for the mispelling. It's funny how most peoples names get shortened, like I am called either Imperfect, or Iam, which both work fine for me, but I wonder how many people get po'd about the shortening of there name?

I have 3 pretty good looking plates of my mushroom tissue sample. Is it a bad idea to make a LI directly with the initial colonized plates that include the dropped piece of mushroom in it?

How would you be able to tell the cleanliness better from the initial tissue and the next one without it? The tissue itself could have something within it or what?

Beats me I do it just to be safe. I'm never in a hurry with stuff, so I don't mind waiting. I'm sure you could use the initial agar plate and be fine though. People drop pins in jars and have them colonize clean...

the clone tissue sample you took may have a spore or something on it that could germinate later so taking a transfer makes most of us feel a bit more safe, the initial plate has potential non-sterile tissue on it. it should be sterile as it's inner flesh but taking a single transfer at least is a good standard procedure

It's just a better safe than sorry kind of thing. I have used my first plate when I was in a rush and it worked out fine but I prefer to make a transfer just to be safe.

Ok, so it's definitely the shit inside the tissue that isn't visible yet? Good to know. I'm only doing 3 quarts and I already have them PC'd so I'll probably just use the tissue sample because I don't care if it contaminates. I have too much spawn right now anyway. Thanks for the help, y'all.
