agar contamination?
    #20995802 - 12/18/14 07:08 PM (9 years, 4 months ago)

Made some agar plates two days ago and starting to see some spots in places it was inoculated in. How can I tell if it's mycelium or bacteria? It's a little clear and small but it looks0 like it has branches coming out.

Two days is pretty fast for inoculation, at least for me

Post a pic!

Don't really have pics cuz it's so small... but yea I know 2 days is fast :/ thats y I'm concerned... it is clear but it looks like it's stringy and growing out like mycelium does. I'll try to get a pic but it's been looking fuzzy through the phone camera and I don't want to open the lid in fears of introducing contaminants.

Is it in the area you i nnoculated? Did you drop spores on or use a loop?

I usually see a clear spot before I see white myc, or when I make a transfer I can see a ring of like tiny clear threads in a circle where the myc is going to grow out. If thats what it is,you'll see myc fast

Keep an eye on it, as soon as you see myc growth transfer a piece to a new plate

Hopefully I'll have mycelium soon!

so i was looking at my agars and they all kinda look the same, they are developing clear dots around where i inoculated the agar. i dropped a drop from a spore syringe in 2 locations on the agar. today is day 3 and it looks like this.

i know 2-3 days time to germinate is very fast so im worried about contamination. what do you all think? i dont think the pics are very clear but i did my best.

so as an update the dish has a bunch of those little clear dots, however they arent so little anymore, but still clear.... about how long until i should have white mycelium? the dishes seem to be growing nicely, the clear indentations seem to be growing like fingers and stretching out  i's not going to need to transfer them am i?

Give it a day or two to thicken up until you can clearly see strands and then transfer.
I normally wait until I can see healthy and clear growth. You should also use food coloring in your agar. Makes it easier to distinguish contams from everything else:
