So I took a look at my agar plates today and the one in which I dropped some gill scrapings from a dried mushy from is certainly contaminated. Looked pretty yuck inside, like a green slime.
On the side walls in the condensation was two white spots, around some pieces of mushroom gill. Not sure whether it is myc or white mold. So I transferred them to either side of a new agar plate. So lets see what happens.
I took a look at the other two plates in which I used a MS syringe, no signs of growth however the smell isn't all too pleasant. Not bad like the one with the mushroom gills but certainly had a smell. Is this normal?
Would the instant mash and/or agar possibly go off or are there contams in there which I just cannot see yet? So far does not seem as if I am having much luck with my agar though

So there is a possibility that the agar with the MS is alright and will one day grow some myc?
Then a very good possibility what I just transferred out of the agar plate with the gill is actually white mold and not myc?

What do you mean the smell of the one you inoculated with a syringe isn't pleasant? Please don't tell me you took the lid off and smelled it. Ita highly unlikely that you could smell a contam before you saw it with agar

Smelled it! Lol!
I smell my clean dishes after I'm done transferring from them, but that's it, and that's only because I'm a fanatic for the mushroom smell.

Mono, bi or tri,
Which petris do you guys use? I bought 40 mono's and I'm gonna buy a box of 500 in a day or two here. The monos are alright but I think I want to be able to section off growth. Using one drop for a whole mono versus three drops per petri sounds a lot more viable.
Thoughts?

Hey, I said I would report back on that transfer, no luck, just more of the contaminate Rhizopus, I do beleive that's what it was, matches every pic. I have ever seen of it, and description too, not a pathogen to man, by itself.
I'm getting ready to make up some MYEA, in Violets screw top containers, I have actually had a lot of luck using those, as I'm sure I would have the mini-rounds, if I had not fucked them up(me doing something stupid, is what fucked them up).

I did. I know, stupid as it allows for a ton of contains to get in and contams loove agar. Just about anything grows on agar.
I got bored and figured it could be a nice way to learn. See what happens and I have some spare agar dishes waiting to go into the PC.
I was curious after transferring from the plate which had some gill scrapings on them. Also heard that contams smell. So I figured "screw it! I've got another busy sitting there. Can always make more. Let me possibly learn the hard way".
Haha
It doesn't smell too pleassnt. I certainly would not want that agar in or around my mouth. Any ideas why? There was nothing growing on top of the agar that I could see. Most bizarre.

I take my lids off I peek and like to pic day by days. I can't stop myself, so I do it as safely as possible without busting out the SAB.

Wait what? Maybe it's because I've been using lab grade stuff as opposed to the asian food flakes? never had this smell though lol.

So you'd rather risk contaminating once clean cultures that took 45mins to an hour to prep than break out a SAB, spray and wait 10 minutes? The mini rounds are clear(ish), at least enough to spot contams, especially with a bright flash light.

Pudgy, it sounds as if you are playing with fire there! :P
Eh, some say it does not stink and others say it does. I do not recall it smelling when I first poured it.
I am not using mini rounds as I cannot find them locally, I found some other cheap Poly prop containers which I thought would be suitable. Probably not as easy to see through as the mini rounds might be though. Blue lids surely don't help and then the condensation on the sides inside don't make it any easier for me but I use my phones flash. It sort of gets the job done.
I just started with agar to see what it is like while I wait for my cakes to colonize really. Figured I might as well get some experience while I wait. Good or bad it is still experience and I would have learnt from it!
I will keep monitoring these agar plates and see what ends up growing.

That's not good... Did you use food coloring? If so what are the chances of you having used some with sugars present? If you're sure there's no sugars in there, there could be some pretty sub-optimal growth, if there is any.
