A week for spores is nothing. I had some where they didn't germ after a month. I then forgot about the plate and found it 4 months later and to my surprise the spores were just starting to germ. I made a few transfers and got a grow from it. A week is nothin.




Oh wow, damn. So syringe or scrapings from a print will generally take quite a while to germinate on agar?

So best is to transfer myc to an agar plate or clone (use a piece of the mushroom flesh)?

However if you got a new cube print and want to get going with that, not much you can do other than just wait?

I have had fresh spores germ on agar in as little as three days. But 3 weeks can be just as common.  Never is also a possibility.  It will happen when it happens. If your paranoid then do up some more, I usually streak three plates or more anyways.

So I've been dropping some APE spores on Agar. All of my other agar work has come out good but for some reason my APE keeps coming up with bacteria. I'm gonna do three or four plates and test all the syringes since I've been using the same one.

Bacteria?

Yeah bacteria :thumbdown: Try germinating them in a grain jar. I had a PE syringe that had bacteria. It germinated on WBS and the bottom of the jar had bacteria. I picked out a piece of myc that was climbing the jar once it had colonized fully and put it on agar then I g2g the top half of the jar to 6 other quarts of grain via a spoon. Worked out for me.

At this point it has to be the syringe. I've only done a handful of Agar work but the only thing that has contamed on me is this APE. Gonna give it another shot with all three syringes tonight.

Just wish I knew what was going on :shrug:

I tried scraping myc off infected BRF and putting them in grains and got bacteria :lol:. I'll shoot up one jar tonight :rockon:.

You are basically taking advantage of the seed soaking up the water. The bacteria isn't going to germinate where there is no water. The spores will germinate on top while the water drips to the bottom. Let them colonize the top fully or I let the whole jar colonize and then I took a piece of myc that was climing the side of the jar and put that on agar. Then I g2g the top half of the jar to 6 quarts and threw the rest out cause there was a visible spot of bacterial contam on the very bottom. But the bacteria isn't going to show up where its dry. Idk maybe you can ask cronicr, he is where I got the idea from.

Bacteria can grow without lots of water, just not as fast. The reason it wasn't as much a problem in brf of grains is because agar and LC are much easier mediums for them to colonize move through and feed on. Rest assured the bacteria was present In the grains or brf if it was in your syringe, just it was simply unable to colonize the media faster than the myc.

Bacteria cannot grow without water. Most bacteria are killed by dry conditions. Some can survive under dry conditions for quite a while however they will not grow or reproduce.

If you think that fully hydrated grains don't have enough water to support bacteria you need to grow more :thumbup:

Bacteria does need water to survive however it will draw all the water it needs from the grains or brf as the case may be. If you don't believe me just take a bacterial wedge and toss it in a grain jar and see what happens. Obviously if the grains are too wet then the bacteria can spread faster, but a shake can move them around with ease as well.

Why do you think i started coloring my agar in the first place? To make contams like bacteria show up easier. Even a tiny amout of bacteria on a wedge can fuck your jar :frown:

Is this plate looking ok? Most of mine look about like this. The part coming off of the tissue looks good to me but there are little clearish white circles around the tissue that I'm worried might be contamination. Here's the best pics I could get.

Those circles are contams, looks like bacteria. Take a transfer from the clean myc coming off the pin real soon.

10% peroxide solution - optional (10ml 3% peroxide diluted with 90ml water)

that's a really funny way to word that. 10% peroxide would eat your hand pretty quick. 1:10 dilution of 3%. you can't even buy 10% over the counter. I get what he means but damn that could be confusing as hell.

I work with 26% peroxide stabilized with peracetic acid(another peroxide) on a daily basis get that shit on your hand it will seriously fuck you up in seconds. it's hard to breathe around that shit when you're pouring it.

that's crazy since you have to dilute it anyway, that's an ultra unsafe concentration to handle. 1.5 ounces of 26% goes into 5 gallons of water. and that's what we use for sanitizer.

that's for the hydro shop 29% stuff.
it doesn't have the peracetic acid in it so it's not going to make you unable to breathe around it but still that's some dangerous stuff for some retard kids trying to grow pot but then again I'm all for being able to buy dangerous shit and if you fuck your shit up that's your own fault for being a tard.

either way at those dilutions the peroxide is less than a half a percent in strength.
