If you use glad wrap. Cut the roll into smaller 2" rolls. It makes life much easier.

OI! I made a Violet-Pasty Plate. I put your agar into a Screw Top ZipLoc container... I meant to have a huge amount of this great mycelium I grew, but I made the agar layer so thick, that it's like thicker than a pizza. So that's stupid. :laugh:



Normally what I do though is pull the wedges, lay them sideways, cut the excess off of the bottom of the wedge and lay it mycelium side down on the receiving plate. I'll probably be trimming the excess when I inoculate with this, or I'll make a LI with it :shrug:

You should not need to trim the excess. That's just more vectors. If it bothers you use less agar per round. Having excess agar is not an issue no matter what you're doing with it. The myc will colonize the excess quickly and the extra nutes will help it to leap off stronger once it does.

I meant excess in terms of depth of the wedge. The mycelium just colonizes the surface if I'm not mistaken. I prefer to have deeper agar plates because of the storage capabilities. I'm not doing slants yet, nor can I afford to, so short term storage of my plates is necessary. You and others have pointed out thinner agar's downside is definitely the storage capabilities.

It colonizes the surface until you cut it or it gets to an edge, then it'll colonize all of it.  The thickness of said wedge will never really matter.

There really isn't a problem with thick agar, if you can get a piece of it loose then you've got what you need.  Only downside is it can be difficult to get a wedge depending on exactly how thick.

I make thicker plates for cultures I plan on hanging onto for a while as well since I haven't figured out slants yet.

I poured 50 plates today without a flow hood, a flow hood is more of a pain in the ass due to the time it takes to set up and how long you have to leave it running to use it.

I'm sure it doesn't resonate well with a lot of people. I suspect that's why some threads/teks/methods are more or less popular, because of the attitude that goes along with it.


I use saran wrap on my dishes but I've got a roll of parafilm on order now that I work in a lab :dancer:

Has anyone tried not snapping the lids on the dishes for the PC? That is, just setting the lid on the dish, then putting the foil over it. I've been doing it since my 2nd or 3rd batch, and its made dealing with the containers a lot easier. I just have to remove the foil, then I can just pick up the lid without any force.

I tried that once with the larger Ziploc ones. It got sucked shut while PCing and got crushed. Never tried with the glad ones but they might indeed be better suited due to the smaller size.
