You can get a good isolate if going from a nice clone in no time at all. I'm doing PE right now as well, but also GT, TC, Creeper and B+. I'd get bored and antsy just doing one thing...damn ADHD.

For crazy isolation potential I would go with either PE, APE, or KSSS. While any cubes have great potential for wacky or excellent things, those are special in my book. But conversations like this should be in the strain thread

YES! God I hope my KSSS mono comes out with some THICK fruits because I think they're BEAUTIFUL and would love to cultivate more of those.

I grew some KSSS years ago and was really impressed by the beauty of it. Hence, why I am doing more and going to refine the strain that I have going!

I know, not a great picture. But this fruit's stem was twisted around like some old bonsai tree! it was so pretty.

Hey Pasty I've got some APE grains almost ready to fruit but I'm concerned that since these spores are hard as fuck to collect that I might have trouble from senescence once I'm done with the set of grows I have coming up. Is there any way I can be sure that I've created a culture from the actual spores of them? I don't want to waste time playing with a dying culture swab accidentally. Or would it just be better to get another syringe?
Also my jar has 1 or 2 grains that look extra moist and aren't colonizing, they're pressed up against the glass in the jar. I've had this issue before and it usually meant bacteria but this jar recovered fine and didn't show any signs. Do you ever see grains like that eventually colonize or am I better off trying to get them out? (it's worth mentioning that the grains in question were like this right out of the PC once the others dried, they never really grew into the wet state, they just never dried all the way after a few shakes.)
Feels like a noob question to ask but these singular wet grains that don't wanna colonize have been a presence in my shit for a year or two now and it's been annoying as fuck.

Use every drop with care. Clone and isolate, you will need to anyway for consistent results. I'm still searching for the keeper so to speak as well. Then slant that shit. Spores are hard to come by so treat them like gold.

I'm not going for isolates just yet, as long as I can somewhat limit my genetics with clones, I'm fine with that until I get better at not spattering everything with bacteria.

I'm sure it's been covered in the millions of posts but can I use 1/2 pint wide mouths I was using for jars for this method?
Or the tall 1/2 pints? Can I use aluminum foil to cover instead of paper towel? Or two coffee filters with a rubberband around them?

Yes you could use the half pints, there are several no pour teks that call for them. The tall ones are a bitch because of their height tho, and they are really bad for condensation. Dealing with rings on lids or threaded lids is also a PITA IMO. They also take up way more space in both the PC and the fridge. Also cost more. But if you already have them then go for it. Trying things is important. You might like em, I just don't.
The paper towel goes under the foil. Most teks skip this but I like it because it keeps the micropore from getting soaked. Coffee filters are kinda useless unless you use them to filter the bigger chunks from your agar maybe, tho I never bother with that.

Wow, Yeah I must have read the paper towel part and asked my question. I've read your tek four times and it hasn't clicked that you're using aluminum foil ON TOP of the paper towel!
Gotcha Pasty, Bought some APE syringes so I'm gonna play around with GT until I get my method down and then put my diabolical plans into motion.

This is the first transfer from another piece of agar. One side is clearly different than the other, but they both grow outward at the same speed. Should I transfer again from the "stronger" looking side or what? Just looking for some info on whether or not I can tell anything from this.

Also that other side appeared shortly after and grew with it. I figured it was just a satellite contam. Yes?

Even the strong side looks a little wispy so I would take from that side if you can. Something about that mycelium looks off to me though.
Man I've been having bacterial spots landing on my plates pretty constantly. I haven't tried out my new SAB yet and that might fix it. I also ordered a torch because flame sterilizing with a fucking candle sucks.
I wonder if there's anything else I'm not thinking of? Should I get a tyvek suit to wear maybe? I already wear a surgical/dust mask thing to diffuse my breath a bit.

I use the Tyvek suit and tape it to my gloves, but I've also just used a thin sweatshirt fresh out of the wash. It seems to work just as well. Muda doesn't use either, just gloves and his arms wiped down with iso ahead of time.
Yes, turtle container. Bought it on accident. It's cool but makes it hard to snap pics without going into SAB and removing the lid.
Um, what looks off about it? And what does wispy mean in this context? Isn't that what mycelium should look like?

The white fan looking white stuff looks like mush mycellium to me, the stuff coming from the center to 12:00 to like 3:00 , but it appears as if you have a mold or something underneath it and all around, I could be wrong, but that's how it's looking to me, I would try to make about 3 transfers from the section I said looks good, try to just scrape a little off the top for each transfers, that is just IMO thought, but I' m with inocuole, the rest just don't look right, but that one section of real white and dense growth looks like what you want.

Hmmm, interesting. The big chunk is just the transfer spot which was way too big a chunk. I'm still new working with the blade. I hope there's no mold there.

More like 11:00 to 3:30 looks good, looks as if there is a good leading section from like 11:00 to I forget where, that you could dig into the agar and get a clean sample, take a look and you should be able to notice what I am talking about, then if you get a good transfer, it should look similar all the way around, that has been IME.

The "weaker" looking mycelium? Can you explain why that's a better transfer spot than the more dense section I outlined?
