You dip it into the non-colonized portion of agar before you cut the mycelium out. It's basically like when people spray out a bit of boiled liquid in the tip of a syringe to have the fresh solution cool off the tip.

propane torch ftw  red hot in seconds
I wrappped the end of the handle in a paper towel and wrapped that in masking tape. Now i never feel the heat.

Too cool the blade you should dip it into the reciving plate to cool. This will prevent cross contamination. Cron has a plate that he designates his cooling plate and just does not transfer anything to it.

I use a blow torch for sterilizing needles/scalpels and it works great...few seconds and it's glowing.

I've seen people say this would disrupt the still air in the room/create currents, which it probably does, but it's never been an issue at all. Just don't have it in your SAB :lol:

idk a propane torch works great. it's outside of the SAB anyway so the air current thing is moot

I have used a propane torch for a while and love it. I ran out of propane last week though and instead of buying a new canister I picked up a smaller butane torch for 20 dollars. It works the same but is a lot smaller and the flame doesn't get my sterile work tent quite as hot. Don't get a torch at a head shop. Go to a hardware store. They are way cheaper there.

I use my gas stove, heat the scalpel then I put it in a bowl of alcohol, heat, use, alcohol. Heat, use, alcohol.

No contam yet, but could be dumb luck.

Also, I've got two of my 3rd transfers showing really ropey growth -- super stoked. Going to cook up some agar now for my 4th transfer plates.

Don't put your instruments in alcohol after flame sterilizing. That's backwards. Once you flame sterilize, your injection or slicing point shouldn't hit alcohol as it just reintroduces your instrument to shitty conditions.

Flame and you're good.

Yup! Heat kills EVERYTHING, alcohol doesn't, so it makes no sense to do both, especially alcohol last. Although I will wipe down instruments with alcohol with parts that can't be flame sterilized, e.g., scalpel handle, syringe plunger, etc.

Updated pictures of some of the last plates:

1) Is this a prime candidate for transferring since it's so scattered in development?



2) Thoughts on this one and it's second cluster of mycelium? I planned on making some more inoculant and LC soon, was thinking of these guys?



3) This is the plate with the mycelium growing on one side, partially on the rim of the plate. I can't tell if this bacteria or just the funny angle of the red agar being eaten away and taken over by white mycelium. The top of it is only white.



Appreciate thoughts on any and all cultures! Thanks!

The satellite is a contam. The others look like they are chock full of genetics, transfer them and see what you get. Might be some ride alongs in there so watch the recovery close.

Are satellites nearly always bacteria or is that one special? I dropped the first plate into 2 LC's just a minute ago, but I'll transfer out the satellite dish (hehe) into new agar when I have some.
