Sterility advice...

Ok. Have a hepa machine rated to a 7x10 ft room in a 5x5...thats been bleached to high hell, with all vents closed and a decent sealing door... planning on doing a liquid to liquid innoculation. Am i missing anything? is there anything else i can do, besides shower?

Are you using the HEPA machine as a flowhood, or is it just running in the room? A breath mask would not hurt, and lots of ISO to wipe things with, I always wipe the jars over with iso especially around the opening. Other than that, Gamma Irradiation perhaps? :smile:

nah, hepa's just running...kinda cleaning the air. Overhead exhaust fan should do flowhood duties....and as far as iso, i think the sheer amount of bleach should probably do...and it'd require a run to the store. As far as gamma irradiation..don't make the spores angry!

Well, reaching into my belt of mycology related ninja tools, and using a 10 CC syring (freshly emptied), i intend to crack the liquid jar, quickly suck up a goodly dose of innoculant (wether it contains visible mycelium or not) and then dose thru a hole in the lid on the other 3 or 4 jars.. There's not many specks of appreciable size, but the solution is diffusing light pretty well, so i assume its ready. Givin the sheer amount of antimicrobial activity and the presence of mycelium and peroxide in the stock jar, i don't anticipate a problem with the jar being open to retrieve the innoculant. The other jars are sitting in the pressure cooker, cooled and awaiting my attention. Any modifications?

Not really. Just loosen the lids a bit (all of them) so you can crack them open with one hand before you do anything. This just helps so you can be quick in cracking the lids AND THEN IMMEDIATELY CLOSING THEM. (This goes for the stock solution, as well.) do everything with one hand, so you can pull the stuff up with the syringe using the other hand (make sure you get the coordination down first.) If they're not completely tightened for like 2 minutes it's not really a contam. problem. Also, it might help to get a flame going so you can flame sterilize the (already-sterilized) syringe between jars.

overhead exhaust fan? Is that filtered air? I like to have all air current reduced as much as possible. Turn off the air cond. and fans. I wouldn't recommend a fan blowing down at you.... unless I am misunderstanding your post.

i sat on my bed room floor and did it.....just clean a area lay down a little table. covered mouth with shirt. stoped all air currents. cleaned hands and tools(right befor and after each step i used them in) slow steady movments. mine turned out great

Overhead ehaust as in it sucks air OUT of the room Did it yesterday, jars already colonizing well. Intend to re-bleach the interior of the room, then take my 8-12 quarts o shtuff back in and use LARGE amounts of water to innoculate. HEPA filter appears to be doing its job...its kinda rated for a much larger room. Got a bleach soaked towel over both vents just to be sure, and all work is being done fresh from the shower....while nekkid. All heckling can begin.....wait for it...WAIT FOR IT>>>.now.

Glovebox?  GLOVEBOX!?!?!?  We dont need no stinking GLOVEBOX!  But no, seriously, if there's anything mycologically or bacteriologically alive in this apartment that can infect my jars, i'm moving out, it can have the place.  It deserves it after what i've put it thru  :eyemouth:

actually the glovebox is the best way to go....
they cost under 20$ to build, last forever, and have 100% sucess rate

they are a bit limiting, but depending on what kind of work you are doing it may not matter....

theory being, why sterlize the ENTIRE ROOM, when u can just sterlize a little box?
no need for filters or Positive pressure system....

just 2 gloves ,a can of lysol, and your ready to rock.

unless your ready to build a flowhood,
get a glovebox...
and NEVER i repeat
NEVER get contam again*
