Boil a quarter cup of whole oats, put them in a glad round mini or "pasty plate", pressure cook it and when it's cool -- drop a piece of your culture into it. I did this -- except I shot it with my syringe, for fun.

My bottle tek is perfect for testing. You can just make one or two bottles for each plate you want to test. So with the amount of grain/sub you would use in one mono you could test up to 18 plates.

That's interesting. Doesn't it require the blender though?

Have you read the updated options in my LI tek? You can probably fide a used oster blender at a thrift shop for a couple dollars or a new one for like $25 or just buy the blade assembly and use a drill.

You mean LI bottles. Im going to try the shake with marbles and glass in the jar method till I get the right bits. So, would you make a clone first of the mono and use that?
Ive seen the updates but im from the UK so need to work out what does the same job.

This is the only growth I have in all of my 8 cups and they're all contams. I'll get rid of these soon and start over.

Isn't it cool you have such a quick indicator of your sterile technique? Make corrections, go again. Results. Make corrections, go again. Results.
Bad ass -- I LOVE agar!

How are you inoculating your agar? I think I saw you post about a plate with a full cc on it before... IME you will never have success with syringe directly to agar. Either squirt the spore solution on a sterilized metal loop then swipe, or swipe spores across the agar. My best results come from clones

Yup, it's neat. I'll record my next inoculation attempts so I can check my technique. I currently ONLY have spore syringes though and like 1CC left in each syringe.
A fellow poster is going to mail me a print though so I'll have that soon enough thankfully.
Anyone good with agar technique who feels like being awesome should upload a video of their inoculation process. You can usually just set your phone on top of the SAB if you have one!

Dude you should ask MonkeyJesusFresco and MudaFuka they had really really good tips. Monkey's house got trich bombed and he said he's had no contams for 3 months because he got his technique down.

I make videos of my sterile work all the time. I like to watch them so I can find things that I may have done wrong and improve my technique. I think everyone should do this. It really helps. I tried to upload one of them but couldn't get it to work.

Do you feel like emailing me the videos? I use YouTube to upload and just put the account on private so only the people I link it to can see them.
If you need help, let me know. I'd love to see a video of you dragging spores across the agar with a loop since that's my next step.

I actually don't have a video right now. They take up way too much space on my phone so I erase them shortly after I have watched them a few times. I have 30 un inoculated plates ready to go so I'll make one this weekend if I have time.
