What exactly does a satellite mean in this context? And yeah, I did a few things wrong during this for sure. I am pretty sure my inoculation technique was awful, I shouldn't have taken off the paper towels above, I didn't have micropore tape, I wiped the lids with an unsterilized paper towel, I probably used too much Karo, etc.

Good thing they're so easy to make! I'll try again soon when I have a spore print and the proper tools.

A satellite colony is a mold colony that pops up away from the inoculation point. Meaning it was introduced by poor procedure in the SAB. mold colonies that are right in at the point of inoculation usually indicate contamed inoculate.

I see. I'm using a syringe that I've already used twice, but have flame sterilized and wiped down each time I put it back in it's bag (and before using). So I probably knocked off some random crap into my plates while handling them out of the PC? Sounds like me. But I ain't no bitch, chef.

I've re-used a bunch of needles and it's never been a problem as long as I flame sterilize.

How would you knock crap in your plates taking them out of the PC? Or did you mean in your SAB?

I think the most important thing to remember is. You want to keep your hands well clear of your open plates.

I just mean from the time they came out of the PC until now, obviously something went wrong since I have mold growing in there. I spray down my SAB with soapy water and use what I believe are pretty sterile techniques when inoculating, but I obviously did something wrong.

I wonder if it could be things falling through the Tyvek/surgical tape crap I used instead of micropore? I'll just record my next inoculation procedure and have people critique it.

That's definitely something I need to work on. I actually am having trouble with that because I made my arm holes way too close together. I'll have to buy another tub to use down the line and make them shoulder length.

Do you inoculate at an angle to avoid your hands going over the plate and to shield the substrate from falling debris?

It can take a while to really get your sterile tek down. I went threw tons of bacteria covered plates before I finally got proficient at doing sterile work. For a while I thought I would never get it right.

Thanks for the words of encouragement. I'll just pay better attention to my hands next time. It'll be easier with a better SAB that allows more movement, I'm sure.

It's super easy to make these little Pasty plates and as we all know, agar tells you quickly if you fucked up, so it'll be a good learning experience, for sure.

Do you think I should PC my 3 contaminated plates or can I just go dump them in the garbage disposal and rinse them out with soap and water?

I have never bothered to PC a contaminated plate or jar. Just don't open them in your grow area. The way I see it. I throw moldy bread and chees in my kitchen garbage so a moldy plate isn't much different.

So I have 6 Pasty Agar dishes.  They are from  4th transfer and a 5th transfer cultures.  Ive had some great results from both when fruiting but now id Like to step it up and make some mono cultures.  Looking at the blurry pictures I dont think im far away.  I could do with some advice on next the next step though. Also Could I take the one with the most strains and make a LI?  Are they big enough yet?

I do it in my kitchen sink. I fill it with hot water and dish soap. I just open them under the water and wash them like normal dishes.

I haven't even had to worry about that much, only had 1 satellite in the last year.

Ed those look good, your really close.Be sure to save each master plate to transfer to a slant as soon as you decide which ones are keepers.

How many monos do you think I could get out of that lot?  Whats the best way to test them.  Frank did mono tubs but thats probably a bit too much for my purposes.  Maybe I should start another thread so we can get back on point......

Thanks, Edmunter. This guide is excellent and I will definitely reference it when I do my next agar session. I wish all Teks were written like this.
