No growth on agar.

I'm trying to get action on agar. I have BGA from mycosupply and platic petri dishes. Neither viable spores or fresh mycel. will expand on the plates.

Does anyone have a good recipe (water to solid) for BGA.

That's the only BG I know of, and it's used for Salmonella  :tongue:

I just went to the site, he means Bacteriology-Grade Agar.  In the mycosupply website, in the 'BGA' section, there is a link to sporeworks that has recipes.  There are also many here at the shroomery, the confusing thing was the 'BGA' term.  Most recipes refer to it only as 'agar'.  It's no surprise if you got no growth using the BGA alone, as it must be supplemented in order to be fung-o-philic.

Yep, Biological grade. I had assumed it had enough myco nutrients in it:
A.)being sold by myco supply
B.)we grew myco (not p.cub.) on it in microbiology lab

Agar by itself is NEVER used as an alone ingredient in mycelium culturing .(except in some very rare cases where one wants to separate certain contaminations from mycelium)

I'm sorry to say but that has to be the dumbest mistake ever. One would think that before one invested money and time into growing shrooms that they would do a little research. I don't know what to say to someone who tries to use agar without adding any nutrients. that's just plain ol dumb......

What are you talking about???
If that was the case every spore syringe shipped would be mycelium by the time it arrived.
About the only thing that is going to happen is you may rehydrate the spores if you scraped the spores off of a sporeprint.
Other than that you will not germinate spores solely on agar..

Lol, no, seriously -- like Anno was talking about above -- if you make water agar you can use it to germinate spores and take the periphery of the mycelium and it should be sterile. This is great if you need to work with a dirty spore print.

I think agar itself contains some nutrition, but very, very little, so not much will grow on it (give it time and shit will, though.) Spores do germinate in water, too, though -- just look at the water from a spore solution under a microscope. You'll see some germinating....

well I dunno, it seems counterintuitive to everything i've learned. however, I must admit that I've never attempted to germinate spores on agar only.

Why? The spores won't all germinate in water, but a few will. Keeping them in the fridge should help to slow this down, but spore syringes do loose potency over time -- I think this is a large factor.

I don't know what you are seeing but I really doubt you could see a spore germinating without a microscope.
