Breeding Contam Resistance?
    #20627531 - 09/27/14 07:26 PM (10 years, 3 months ago)

I had a thought which is either genius or heroically moronic, looking for some feedback. Theoretical tek:

Hydrate your WBS or grain of choice as normal. Load it into jars, but leave the lids off. Then, contaminate the ever living fuck out of it. Spit on it, get your fingers all over it, rub it on the toilet seat, stick it in the refrigerator, whatever.

Now, let the contams colonize the jar. Bear with me, this will make sense soon.

Now, once the jars are fully colonized by a rainbow of contaminants, throw the lids on and PC them dead. This step might take more time than a normal PC, but 8 hours at 15PSI is bound to kill anything.

Now, inoculate your sterile jars with a multispore syringe or LC prepared with multispore.

Growth would probably be slow, but in doing so you would be specifically selecting for genetics that were capable of digesting the proteins of common contams and growing in the presence of any defensive chemicals excreted by said contams. From there you could either isolate the most successful strains via clone to agar or print and repeat the process over multiple generations, to yield a strain that is at least slightly more resistant to contamination.

Thoughts? I know this wouldn't make a magically contaminant proof strain, but I imagine it could be a useful selective breeding technique. Alternately you could do the same on agar, contaminating plates before remelting the agar, running it through a blender, sterilizing and repouring the recycled agar.

Seems like a waste if all you are doing is looking for a cube strain that is "contam resistant".....they all are, if they fully colonize the substrate you put it on.

Fully colonized = contam resistant.....any cube can do this.....

It would be great if someone made a GMO cube that is contam resistant
