I'll upload some microscopy of that plate with the spores germination. I made 5 plates and they're all clean so I figures I could butcher one for science

I tried a multispore wood shaving tub. I basically just added pasterized wood shavings to my previous tub that was loaded with spores, I got zero germination, so it is definitely a good idea to start them on agar first. It will be easiest with the G. resinaceum group.  Other species, G.tsugae in particular are near impossible to germinate.

What is really strange is calcium oxalate(my assumption) crystals are everywhere when I took a look at the mycelium. You'll see in a few hours when I upload but its like the mycelium sequesters Ca and did it itself

so the pictures are kind of shitty. I was in a rush. I did use 10% KOH solution to try to make some nicer looking pictures but :shrug: I didn't really take my time with it.

I couldn't find any spore germinating or spore with hyphae coming out of it. I got close but I'll have to try again later to get a real good picture of a spore in germination.

I couldn't find any clamp connections either. I know not every species of mushroom makes them but I may have just not seen any either. Does any one else know if the ganoderma species make clamp connections?

Yes Ganoderma make clamp connections, they can be hard to observe at times though. Calcium oxalate, would that be diamond shaped crystals? I get those frequently but never really thought about what they might be.

yea if you look up Ca Oxilate pics on google. but I see oxilate all the time in other things and it looks exactly the same, but i have never seen it on agar plate scrapings, or ever on YPD for that matter which leads me to believe it's the resinaceium itself causing that?

Could be, to be honest I haven't paid much attention, but I do see them when I am looking for chlamydospores, which I typically only look for if I suspect it is in the resinaceum group, maybe I will start paying more attention. Thanks for solving the mystery of what those are! :thumbup:

All my strains are a little too grown out for microscopy at the moment.  I think I will do a comparative growth set to see if they form with other species, I kind of just assumed they were something in the agar precipitating. But haven't been looking at agar plates under the microscope enough.

So the only thing in common is the mushroom. I just checked an old MEA plate that nothing ever grew on, no crystals to be found, if it was something in the agar it has had plenty of time to precipitate.  I think it is caused by the mushroom.

perhaps. It's very insoluble in water so coming from the agar itself seems really strange. and I've never seen it on anything I've taken from a YPD plate before.

I found this study when I looked up Calcium oxalate and Ganoderma, it would appear there are several wood rotting mushrooms that produce it on agar.
