Issue with Slants, they boiled off or something.

Ok i just recently bought new slants, the 2 from the left.



i prepared 8 slants, 6 with a wood stick and 2 without the wood stick, i wrap them in foil, place them in a mason jar, the mason jar with foil as the lid. When i took them out of the PC the agar somehow got out and end up in the bottom of the mason jars, like this.



The only ones that survived were the ones without the wood stick.



All the other 6 slants with the stick:



I never had a problem with the slants with the screw lids. Any ideas why this happen? why only the ones without the wood stick survived?

Was there water in the mason jar? If I don't have water up to the agar line i get boil over. I think this happens as it cools because the air around the agar cools faster than the water or something like that.

When I did slants, i put the tubes in jars but also put water in the jar and I didn't get boil over.

Nope, the mason jars didn't have water, but then why only the 2 with the sticks survived :confused: ?

I never had a issue with my old slants with the screw lids because i screw them tight and i never have to put water in the mason jars. Next time i will.

It's hard to say why sometimes some shit works and sometimes it don't:shrug:

Yeah, the screw-top is probably why you didn't have a problem before.

I just ordered a 10 pack of screw top Pyrex test tubes to give slants a whirl. I have Frank's tek bookmarked. Good to see other folks doing it and learning from their experiences lol.

They are too full. Seems that if you fill em too full they will start to boil out when the pressure increases. Then it just don't stop. Water don't hurt either.

This slant was 2/5ths full with water in the jar.

lol this slants i bought are pretend to be poured in front of the FH, this is what the guy told me

"These culture tubes are vented so they will not seal.

When I make my slants, I pre-sterilize the tubes and lids in the PC along with the agar. I pour them after the PC cycle in the laminar flow.

As you have seen, when using vented tubes, if the  agar cools too fast it will boil over. When using sealed test tubes you are risking breaking the tubes when pre-pouring them before the PC cycle.

The gap between the plastic cap and the glass tube is meant to be wrapped in Parafilm after the tubes have been poured."

I will try to pour those things, that way i can pour more agar in each slant.
