Agar/Sectoring Questions!
    #20580243 - 09/17/14 04:14 PM (10 years, 3 months ago)

Hey guys, I want to start by saying thanks in advance to anyone who answers and tries to help me out! So basically, I started to do some Agar work with some homemade petri dishes (may make a tek.), I made 5 and have done one transfer. Petri dishes worked fine, i tried a few designs and I like this the best, but anyways I posted some pics for you guys to check out. I have a couple general questions, and a picture specific question. :mushroom2:

1) What EXACTLY is "sectoring"?

2) How long should a petri take to colonize?

3) How do I decide what "sector"/piece of "sector" is the best for further isolation?

4) Of PICS # 1 - 5 which one would be the best growth to use for further isolation? (PICS # 1, 3 & 5 have been the fastest so far)

PIC # 1:

ISOLATION PIC:

In the above picture, which point would be the best area to take for isolation I marked a few for different reasons:
Point A: because the rhizo growth is thicker there?
Point B: because it's more uniform but attached to Point A?
Point C: because it looks more uniform/thick?
Point D: because it's further out and attached to Point C?
Point E/F: would they be very similar to points C/D because they're in the same "sector"? If i'm right about what sectors are lol.

SECTORING PIC:
