About to start working with agar.

Here's what I've got. I know the SAB is too small, but it was the biggest tote I could find at walmart. I have:
SAB Agar agar Potato flakes Light corn syrup and honey (any recommendations on which is better?) Food coloring Makeshift Petri dishes Gloves Tyvek suit Micropore tape All necessary cleaning/sanitizing supplies Propane soldering torch Last but not least, a syringe of PE uncut
I was planning on following pastywhyte's easy agar tek. It seems very beginner friendly. It will be about 1.5 hours before I get started. Any last minute advice?

honey is naturally anti fungal but people use it all the time with success. Corn syrup is a pretty standard source of dextrose.

See that's what I thought, hence the name potato dextrose agar. Never realized honey was antifungal. Kinda defeats the purpose of using it. Also what kind of sugar is honey, anyway?

Shit I just realized I lost my micropore tape. I don't really want to go to the store again. Can I use really tightly stuffed polyfill instead of micropore tape?

If I may give a suggestion
NEVER put your SAB on the floor, put it on a shelf somewhere, close their hand holes when not in use don't move it too much around
you preferably want it in its own room
stay clean, clean tables, don't work with shrooms when you are dirty in clothes(take a bath if needed)
close doors,turn off fans when you do mycology work
else looks good!
read up on slants that is essential if you want to store your cultures you can't store petri cultures as they dry out within a few weeks usually (unless you add wood into them later)
make a slant and you can keep your isolate indefinitely
but sometimes it is preferable to start over anyway.... having a clean isolate can just be very nice
make some LCs too if you want, but be extremely careful about sterility there, so easy to get them contamed
To use sab: spray down SAB with alcohol/bleach, wait some mins gloves on hands, clean with alcohol heat needle red hot outside with alcohol lamp , bring it inside re-heat needle between each jar (is what I usually do to avoid cross contamination) , some do every 2-3 jars, but I always do between each jar
(an alternative agar recipe I love is oatmeal agar, perfect grow media for me, I make a bucket at a time and freeze it, but haven't tested it after freezing , hope it still works... it can be hard to make that is why I freezed it)

Nice. So the SAB isn't too small 
I had planned on showering, changing clothes, and donning the tyvek suit once my Petris are in the PC. I tried making LC once and it failed miserably. I was thinking of making LI with some agar wedges though. My only real question now is if I can drill a small hole in the lids and stuff it super tight with polyfill. I've been drinking and a run to the store is out of the question. Oh and the SAB is on a big dining table.

never made pasty plates before just use regular petri dishes and pour them myself so IDK about the tape.
You don't need to wipe the SAB with alcohol or bleach just water or very diluted soap water works. the still air is what keeps you sterile not the bleach or alcohol.
you can store your SAB on the floor just don't use it on the floor. I use my SAB to store mycology stuff when I'm not using it as a SAB. Like I said the still air and your technique/skill are what make or break your work not silly shit like bleach alcohol and obnoxious precautions.

True, I store on floor too, I have just had bad luck recently
But probably because I inject near a leaky door, door sealing is not good and air from outside is leaking in at times. Or because of broken PC seal, it is partially broken/leaks now.
I have had success 4 out of 4 times (19 out of 20 pftek jars) with just normal soap+water like you say. Many swear to soapwater cleaning
I did it not to get rid of contams like you say, just very fast, to get rid of the nasty stuff I might have dropped from previous innoculatiosn (like old LC stuff spilled on bottom or such), quick cleaning of bottom and lid of SAB, always turned out fine.
Floor is just nasty for mycology stuff, you bring nasty stuff from the floor onto your table when you move it. Lots of spores. But it can work if you are very careful and not bring them in with your arms.

So do I even need to alter the lids for GE? The Rubbermaid things I bought definitely have an imperfect seal.

if thats the case then you deft need to seal it somehow. the tek i stated used screw-top containers.
do you have glad cling wrap?? that would work.

Yes but that is V tek which uses threaded containers. You do need a hole for pop tops (I can't tell from the pic if yours are or not tho), you can use polyfil and while your SAB is small, it will be fine for culture work. You might want a bigger one for G2G but for now your in business

Poly works good as one said. The SAB is much too small IMO, but it will work.
You should get a bigger one, one where you can innoc 4-6+ jars in easily IMO
It's less than half the size of mine, I use a normal bedroller , and added another one on top + made a seal on them.

Yeah that's what I figured. To me it seems like agar kinda eliminates the need for G2G. I mean 6 quart jars fit comfortably in my SAB, and since I'm only growing for personal consumption I see no reason to have an enormous SAB.
So that's awesome. It's your tek I'm following and if you say poly will work fine then I'm golden. I'll keep updating this thread with my progress. Hopefully this can be my agar to grain to 66qt mono grow log. I did spawn a 66qt mono yesterday and it's rockin away, but this grow will really be my first step into doing it right

Once I've got agar down I'm def investing in a bigger SAB. I made a big error by going to walmart instead of lowes. I've been looking all over for a big pack of glad mini rounds and by the time I went through the hassle of going to walmart, I didn't want to go anywhere else.

ive bought 110qt tubs at walmart and 66t tubs and all kinds of sizes i see there, and thats at walmarts all across my state where ive lived.
maybe they were just out at the time. not that you cant use that but eventually you will wanna G2G its so much easier to innoc one qt with a huge wedge like half a plate then in ten days or so g2g it out to 7-10 more jars

Honey loses most of its anti microbial properties after being exposed to high temperatures like the ones produced during the PC cycle. I used it for a while before I found dextrose and it works fine.

Cool. My WBS just finished up so I'm gonna start mixing agar. I know it's 2g agar:100ml water, but I'm housesitting right now and my scale is at my place. What's a good ratio for agar:water:potato flakes:corn syrup? Pasty's tek doesn't go into that much detail. All I have is measuring spoons, but I can get my scale if necessary.

Mixing it up now. Man I feel like an idiot because the info was there all along. Still, using my scale brought me peace of mind. Agar is on the stove right now. Decided to dye it blue because it just looked so fuckin cool.

Well, I inoculated 2 plates of PE uncut this morning. Think I did everything right. Do spores germinate any quicker on agar or is visible growth a week out?

Spores usually germ quicker on agar IME. Its a perfect environment for germination of anything. But like any sterile media germination is highly dependent on the spores and how old they are, genetics, etc.

Neat. After reading more I think my agar is a little deep. I filled up the plates to about 3/8" deep. As long as everything is sterile this won't be a problem, right? It will just take longer to colonize? I have a tub in it's spawn run and another 6qts of WBS colonizing right now, so I'm in no hurry to do anything with the PE. I was thinking of making liquid inoculant with one plate and storing the other one long term. How long will a colonized agar plate keep in the fridge, anyway?

If the inoculate or transferred wedges are clean it won't be a problem. It will make it very hard to isolate from bacteria tho. If you put it in a ziploc bag it can keep in the fridge for a few months maybe more.

Well, let's just hope I did everything with the SAB right. I misted the inside with soap and water, sanitized my hands and arms, sanitized my gloves, wore tyvek sleeves, wiped the outside of the plates down with alcohol, and used a fresh sterile needle, only opening the lids enough to stick the needle in. As long as my syringe was free of bacteria everything should hopefully be fine I think. I didn't see any need to flame a needle in sterile packaging that I opened inside the SAB. Hope that wasn't a mistake.

I have used plates after a few months of storage in the fridge and they showed strong growth right away. I think the fact that you poured your plates a bit thick should help them to store longer without drying out.

Fuck yeah. I was thinking about making some liquid inoculant using your tek since my blender fits pint jars. Does that keep longer since it's liquid? I have some syringe filter and SHIP lids that I got for LC, but every attempt of mine at making LC failed miserably. The LI like you make it seems really awesome. I figure by the time this PE is ready to go I'll have a few more grows under my belt. Kind of off topic, but does anyone know how common albino fruits are with PE uncut? I'd love to make an isolate from one.

I have never tried storing LI I use it the second I make it. If you want long term storage make slants.

I've read a little about slants, but that seems tricky as hell, keeping the test tube at a 45 degree angle and pouring freshly PC'd agar into them. Do you need a special breathable plug for the test tubes, or is the air in them enough?

No you pre pour the slant then PC with the lids on loose. Then pull outta the PC hot and in the SAB remove from the jar or whatever you PC'd them in and let cool on a angle. Its easier than it sounds.

That's why I just use petries and every few months I take them out and make transfers from the ones I want to keep growing and just toss out the rest. I get really frustrated and overwhelmed when I have too many things going on so I'm really trying to limit myself to no more than 4 or 5 cultures going at a time.

Looks like there will be some test tubes in my future. I suppose I'll try making some slants when I eventually grow this PE out. If I get some nice albino fruits I'll definitely want to clone one and keep it around. Man this site is fucking great. I really didn't expect everyone to be so supportive and encouraging. I can't believe how much I've learned in the short time I've spent here. I'm so excited to see some good results. I definitely need to make a much larger SAB pretty soon

Yep its pretty damn easy. People always blow agar outta proportion in their heads and use that as the excuse not to use it. I like the screwtop slants like this one best.

Hell yeah. Looking at that pic makes me want to get into edibles/medicinals as well. Having just set foot in the wonderful world of agar I have a way to go, but I think that once I have a few PE grows under my belt and have made some good isolates I'll get into slants, woodlovers, and edibles. Next year I hope to do some outdoor azurescens and morel beds and maybe do a sclerotia producing species.

So almost a week later I'm starting to wonder if I fucked something up. My 4 pasty plates have just been sitting on the shelf. 3 of them look like growth is starting because I see little pinpoints of white on them, but one of them shows no growth at all.
Since tomorrow morning will be the one week mark I'm wondering if I did something wrong. I'm inclined to think I didn't use enough corn syrup. I think it was pasty that told me that spores germinate marginally quicker on agar, but I've also heard PE takes longer in general.
I'm somewhat inclined to scrap the 3 of my plates that show the least growth and starting them over, letting the 4th one just chill to see what happens. Is it worth it to start over and try again with more corn syrup, or should I give all of them a few more days?
I also think I might have botched the recipe since I was drinking when I cooked the agar up. I still have plenty of material left. My recipe was 2g of agar agar, a heaping teaspoon of potato flakes, 100ml of water, and about 1/4 tsp of light corn syrup, and a few drops of blue food coloring. There's still lots of condensation in my plates as well. Which should have been gone after a few days from everything I've read.
Would someone be kind enough to clue me in on a few things? 1. Did I fuck up the recipe? 2. Is a week with very little growth out of the ordinary?

You used a bit more poyato flake and and corn syrup than i would with a batch that small but it should still work fine. Dont scrap anthing just yet. Nature doesn't have a set schedule. Some times it can take longer than expected for spores to germinate. You should start another set of plates while you wait. Just incase they turn out to be fucked.

If you see growth why would you scrap those? I would wait it out a bit longer..
Muda's right, make some more plates while you wait. I just started with agar as well and it took me a few dozen plates until I got good growth on a few..then those few transferred over to more, etc etc. 4 plates is very little! Keep trying.

I generally make between 20 and 30 plates in each batch. I like to take multiple transfers from each plate just in case some of them contaminate. I usually wind up throwing out a lot of clean plates in the end but I like the piece of mind.
