Agar Questions

1. I ordered Potato Dextrose Agar. The mycellium can grow on dextrose, so I have no need for Malt Extract, right?
2. After mycellium colonizes the agar, how would I transfer it to, say, a popcorn medium?

1. If the Agar you ordered is already mixed then no you don't need to add anything.
3. Cut it into wedges like a cake with a sterile knife, then lift a slice with the point of the knife and squiftly drop it into the popcorn jar. Shake. Best done in a flowhood, or just in a clean draft free room.

That sounds like a huge contam risk. There is no cleaner way to do it than just opening the jars thus exposing them to contaminants?

Can't think of a cleaner way. Working with agar is tricky because of the contam risk, you need good asceptic technique. There are methods using peroxide that reduce the risk. In any case you have to accept that some will fail due to contams.
If you want to work with agar a lot best build a flowhood or glovebox big enough to work with jars. The genius of the PF Tek is the sterile technique IMO, it will has exceptional resistance to contams in the environment.

I've had plenty of success using agar without a glovebox or a flowhood although I would recommend investing in either.
Add some hydrogen peroxide to your agar if you plan on cloning just to be on the safe side. Hydrogen peroxide decomposes at 140 degrees fahrenheit so you need to let the temperature of the agar drop below that before you add the peroxide otherwise it will decompose into water and oxygen and be effectively useless.
