Jars not colonizing
    #2046815 - 10/27/03 10:01 AM (13 years, 1 month ago)

Hello fellow growers!
I have had an odd problem for the last three times i've been making Cubensis-cakes. They just won't grow!

I made perfect cakes in the first time, but now i'm baffled.
I use basic PF-tek with vermiculite and BRF. There is no sterilisation-problem, haven't had any contams.

The second time making cakes, i spotted a small mycelium colony in only one of the cakes, but it had stopped growing. the third and the fourth time i saw no growth in the jars.

I've been waiting over 30 days each time. i've kept temperatures between 23C(73F) and 28 C (82.4F)

Please help me if you have had any similiar problems and found out the solution. My current quess is that my vermiculite is in too big chunks? is there a possibility that mycelium needs smaller size of vermiculite to start growing?

here's more info
sterilisation method used: a normal pot with a lid on, kept 1.5h in steaming water. jars cooled down 12 hours.
spore print: shayanashop, thai/ecuador cubensis.

Please, any info is _highly_ appreciated. I don't wanna make another set of jars until i have some idea why did i fail three times a row.

did you add water to your jars? also have you used the same spores each time? if so there is a chance that the spores are very weak. the vermiculite size is not really that important. other than that I dont know. keep trying.

read the pf tek again.

ur spores may be junk.

how are you innoculating? are you making a syringe from ur print. if so what sterilization do you have.

I didn't add water to the jars when i first got succesful jars, so i thought that isn't so important. was i wrong?
No, i haven't used same spores each time, had to get a new one. how common it is to have bad spores?

when making a syringe, i use a disinfectant to wipe the shot glass clean and let it dry. then i spray some water from the syringe to the shotglass and scrape the spores with a sterilized knife. then some more water and back up to the syringe.
when inoculating, i sterilize the needle after every jar with a papertowel soaked in disinfectant, and after that with a lighter.

Do you really mean you added no water, at all? Or do you mean you didn't add extra water?

1. I mix water in vermiculite. i stop when little water tilts from the bottom.
2. I add BRF to the vermiculite (used a coffeebean grinder to powderize normal brown rice) and mix them well.
3. Then i measure the correct amount of mixture to each jar without tapping it down and seal them up with three layers of foil.
4. Next, i steam the jars 1,5 hours in a normal pot with a lid, very low temperature, just enough for the water to evaporate.
5. after 12h+ of cooling at room temperature, jars we're inoculated.

im not very experienced so dont trust me on this but, i read somewhere that regular brown rice sometimes contains fungicide on it. the place where i read that (sorry i dont remember) said to go to an organic food store and buy brown rice flour. this way you dont have to grind it and also it wont contain fungicide because its from an organic food store.
