Poor man's self healing innoc spot

Nothing too new here, but I thought I'd share. Being the poor bastard that I am, I was looking for a way to make a self healing innoculation/extraction point for a honey tek jar I'm doing. I substituted polyfill for a regular old rubber eraser, super glued to the lid. The hole is underneath the eraser. I'm going to germinate some spores in Karo, only thing is it is Karo dark corn syrup, not clear, it should still be fine, right? Anyways, I'm going to go PC this and some jars, I'll let you know how it turns out.

Well, I thought the Karo might be a lil shaky to test my tek out on, so I switched it to Mickey's Ice. That's right, I'm using the beer tek this time around!

What is the "beer tek" ? Where can I read more about that tek ?

I actually found a reference to it while searching for more info about karo. Apparently beer contains malodextrin(sp?) a type of sugar found in malt beverages (beer being one). Basically, instead of using karo or honey, you use beer. But first you got to cook it for a while to evaporate all of the alcohol out. You can then use it to germinate spores in, and make mycacelium syringes, which colonize faster and allow you to innoculate more jars from a spore syringe, which is economical.

I know that i found this on nans , but the best answer that i have found is good old high temp silicon sealer , great injection ports . I dimple my lids a tad n fill with the silicon sealer , let set 24 hours and is the best.

Have you ever successfully used the beer tek ? If yes, how did you prepare it ? I mean what kind of beer, how much beer in how much water etc. This topic is very interesting to me, because here we have a lot of old beer standing around ;-)

I'm not doing this as perhaps the BEST way to go about sealing it, but it does seem like it should do the trick effectively, and I wanted to try this for novelty's sake. This is my first time doing beer, I'll let you know how it all turns out tommorow how the jar and "seal" held up during PCing. Then I'll innoculate it and tell you how growth is doing. I finally got a heater for my incubator again, so I'll be able to get steady temps of 86 so that temp fluctuations aren't a factor in how it does, since temp will hopefully be optimal. I'll keep you posted.

Well, I PC'd the beer last night, twice. The first time I got it up to PC temps for about 5 minutes, and had to leave. When I returned, I got the PC up to temp again and PC'd for about 20 minutes. The beer looked a lil bit darker, kinda like piss when you're not healthy, but didn't turn brown or black (bad, cause carmelization has occured, doesn't make for hospitable shroom growing environment) However, there was a bunch of crap that settled to the bottom of the jar. Don't know exactly what it is, but I pressed forward with innoculating it anyway. As I try to wiggle the needle around to find the hole underneath the eraser, the glue breaks on one of the edges.
I push the eraser back into contact with the lid before the seal could be compromised, but it seems like super glue and metal don't bind well, next time I'll recommend/try scratching the lid up a little so that the glue can get a better hold on the metal. Anyway, I shot up the beer with ~ 1 cc of PES Hawiian spores and covered the whole shebang up and put it in my incubator, which was at about 78 degrees F when I put it in. Needless to say, I cranked the heat up more, and hopefully it'll be in the 80's when I check it again. I'll update in a few days when/if any growth starts.

The stuff floating in the beer is probably normal.
This is why people recomend karo. It is totally clear. Anything that isn't white is easily spotted.

Hi,
when I was at my med's place I have seen that they were dumping their infusions glasses. They consist out of autoclavable glass and are closed with autoclavable rubber lids that are self healing. There is an alu ring around the rubber, that can easily be removed. I think they would be great for liquid innoculation. They ship in 50 ml, 250 ml and 500 ml sizes. Here are pictures of them:

Sorry to ressurect, but where can I find those med bottles with the sealing tops? I'm a huge fan of liquid culture and that would make my day.

you can buy jars of "sterile water for injecting" at any med supply store... not sure if you need to be a doc, i don't think so...

nice one i have made inoc spots in the past by puting a large glob of silicone bath sealant either side of the hole works well and one tube will brbably do you a few hundered patches
