Experimental Inoculation W/Pics

I went ahead and took it upon myself to do a little experiment in mycology, since I am a noob in training, what could it hurt? 11 days ago when a friend and I did the bulk to spawn step that our grow kits came with (see my first grow post for more info on this), I purposely took some of the rye spawn out of each of the half gallon jars they came with; put them in a bag, and stored them in the fridge... I had an idea in my head... If I could save some of the fungus on the rye berries, I could probably transplant it to other jars and grow more of it (kind of like G2G but at the time I had no idea if it would work). Later on that week, I went ahead and got the supplies required to make 10 half pint BRF cakes. The following was what I did with the fungus in a step by step tek (I wouldn't recommend doing this yourself because your stuff will probably get contaminated, this is just an experiment).
1. Made the standard BRF cake mixture 2. Drilled standard inoculation holes in my 1/2 jars 3. Filled my jars with the BRF mixture (leaving 1/2 inch at top) 4. Fill 1 full pint jar with dry vermiculite 5. Cover all the jars with foil 6. PC at 15psi for 60 min 7. let cool overnight 8. Shut off HEPA in the house to stop any moving air 9. Made sterile working area 10. Sterilized myself (mask, gloves, alc) 11. Proceed to remove jars from PC 12. Quickly and carefully, remove lid from each BRF jar, add spawn to top; then add dry layer of vermiculite and seal. 13. Let colonize
Mind you I worked very quickly and carefully trying to expose my sterilized materials to as little open air as possible. It has now been several days since I did this inoculation, and all 10 jars have growth. I have no idea if they will make it all the way, but I guess we'll see won't we? Here are a couple pics of the progress...


I know a lot of you would frown upon something like this, but it was all I had to work with; and at the time, I didn't know any better.

Well, I had a feeling that our kits may fail. So, at the time, saving the spawn at any cost seemed like a good idea.

What was your procedure for taking the rye out of the original jars? I've never done a G2G but I know it's more contamination prone than say injecting spores, so I'd say contams are likely if you haphazardly transferred and stored your grains. I'm a rank amateur though, good luck!

I did it by hand, with disinfected gloves and working area.
Edit: Sorry, I completely gaped half your question. To answer, I simply took the colonized berries out of the half gallon jars and quickly put them into a brand new zip-lock bag. I stored them in a clean place in the fridge until I got materials to do cakes. Probably wasn't the best procedure. I thought about it after, if I would have sterilized another jar, I could have just put them in there. But hey, I guess that's the way experiments go.

You would have been better off leaving the grains in the original container for storage and then pouring them in to your cake jars.
If you have a filter, like SFD's for instance, you can skip the dry vermiculite barrier.
Do you have GE holes in your lids?

Wait scratch that. I just searched what a GE hole was and NO I don't have them. I just drilled holes in the lid like the PF tek said to do.

Update: These jars seem to be doing well. I tossed two of them because one had some green growth spot and another one I thought just wasn't doing anything, I couldn't see white growth... Turns out it was growing in the center. That was two days ago, the other 8 are all white. Here are some pics. Also, I realized they are only 125ML (1/2 of 1/2 pt) jars, I guess we have some mini cakes. I was wondering why I had so much BRF mix left over...

Yea those are 4oz jars, i used some in a pinch. IMO they won't really produce cakes big enough to get good fruits off.
I had to go to like 18 stores to find 1/2 pint wide mouth jars, but they are so worth it.
I think you did the right thing trying to save some myc  DSP

Well, kit 1# ended up succumbing to trich as well. I put it out in a bush that seemed like a good growing spot for them. Wen't back yesterday to water and a coon or something had a nice mess made of where I buried it; must have been a good lunch.
