Experience with long-term storage in distilled water?
    #2028127 - 10/21/03 10:28 AM (14 years, 1 month ago)

Hello,

I have found a description on how to store mushroom mycelium in distilled water for a longer period of time. If anybody here has done this I would be very interested in his/her experiences.

Cheers,

Wow! I never thought mycelium would live very long in DI Water.

It works good, most of the cultures still alive after more than 2 years.

What strains have you stored in the distilled water ?

I have stored various oyster mushrooms, reishi, enoki, and psilocybe. With the exception of the oyster mushrooms, Everything holds well for at least two years.

Wow, sounds good. What's the problem with the oyster mushrooms? Did you notice any difference in growth before and after the long-term storage ?

An excerpt from said storage link:
" About six milliliters of sterile distilled water is pipetted aseptically into a freshly growing culture. The fragments of hyphae are dislodged by lightly scraping the aerial growth with the same pipette, and the resulting suspension is withdrawn and transferred to a sterile glass vial. "
SAY WHAT?
:oogle:
Can anyone break this down in Layman's terms?
Could this work with the Karo tek using distilled water?
(inject spores in Karo water mix, incubate for growth, seal lid and store?)

This means that you sprinkle 6 mililiter of distilled water over the mycelium on a agar plate, scrape a little on the surface of the plate and suck the so gained water/mycelium mixture up into your pipette/syring. The you can transfer the water/mycelium mixture into a test tube.

No, I do not think so. Putting the mycelium into distilled water lets them fall into a dormant state, whereas it does not fall into this state when you add some nutrients like karo to it.

Another way to do it that I've found easy is to use sterilized wooden handled swabs [pre-sterilized or PC'd] to swab up mycelium. Then dislodge the mycelium into a tube of sterile dH2O. Disadvantage is that you can get pieces of agar in there, but if things are clean it's not a problem. The basic idea really does work, I still can work with a cyanescens clone from 3 years ago, stored at room temp. As others said, not all species take to it well.

Does anybody know how long spores stay viable in distilled water ?

On a spore print for many, many years. don't think they would survive longer in destilled water...

I wonder if adding a little h2o2 to the dh2o would help
out in the early stages, or if you could store an agar wedge
in a jar of dh2o/h2o2..

You can, yes.

You can also store a wedge in a small plastic baggie.
Place it in several layers of plastic so it doesn?t dry out.

What is the longest time you have stored a spore print before you have used it successfully?
