Sanitation question on still air box and jar/lids

1) Do you sanitize the jar and lids before you add the substrate? Or does it not really matter since you're gonna boil it afterward anyway?

2) I'm confused about the still air box procedure. The RR video just shows the guy with a glove on plus some kinda medical sleeve. Do you need to also wear that sleeve he's wearing, or will gloves suffice? And do you add the jars + syringe inside the still air box before or after you aerosol it? In the RR video, they don't aerosol at all. They just go right at it, but in another video, I've seen them aerosol or bleach spray the box with the syringe and jars inside it.

3) Once you inoculate, do you remove the jars from the still air box and transfer them to another box? Or do you just leave them inside the still air box? The RR vid completely skips that part. They go straight from inoculation to birthing lulz.

It doesn't matter because you're going to sterilize in the PC / pot.

2)You're going to get varying answers on this. Personally I shower and scrub my forearms down really well with a stone or luffa. Then I spray bleach-water into the chamber to push contaminants against the walls then go to work immediately afterward. I don't wear tyvek sleeves. Just nitrile / latex gloves and I sanitize them with 70% iso as I work.  I sanitize the sterile media jars with alcohol before I put them in the chamber and then I spray more bleach water and then while I'm in the chamber I sanitize the inoculation points with alcohol again between each jar.

3) After inoculation you can put your jars wherever you want. Just try to keep em off the floor. A lot of people put them inside a separate tote.  I put some on my top shelf of my closet and some in a plastic tote.

Both seem to colonize at the same rates. The place doesn't really matter as long as it's about room temperature and get's minimal ambient light.  I feel like the temperature is slightly higher inside a tote though.

These are good questions that really should be addressed better in the video.

Will 99% iso work just as well?

Also, how do you sterilize the gloves and jars with alcohol? Do you use a cotton swab? Some kinda fabric? Or do you just pour it directly and start lathering it all over?

You also mention you spray more bleach water. Where are you spraying it? Do you mean you spray more bleach water AFTER you insert the jars and syringe into the still air box?

And what do you mean you sterilize the "inoculation points?" Do you mean the holes you poked into the lid? You just run a swab of alcohol on the rim of it?

What happens if aerosol or bleach water gets inside the cake? I also wondered about this as well.

I keep a half pint jar filled with a half inch of iso and a couple folded paper towels for quick sanitation. Please understand that alcohol does not sterilize, it only sanitizes. I use the iso soaked paper towel to wipe down every nook and cranny of the jars and my gloves and everything I place into the SAB.

Yes. The holes you made in the lid should be covered with tape when you pressure cook or steam sterilize the jars.  When you go to inoculate you're going to insert the syringe right through that tape.

Yes. Just an iso soaked paper towel wiped over the tape-covered-holes that you're going to use for inoculation.

I wouldn't know since I've never been so bleach or lysol crazy to get that shit into my inoculation holes / cake.  This will probably never happen unless your SAB is literally dripping with lysol when you go to inoculate.

That being said. Mutation and other kinds of 'contamination' can happen when your mycelium is exposed to disinfectants.  You can search for this very topic to see the results.

But again it's unlikely to happen unless you're going bat shit with the lysol.

Don't forget to flame sterilize your needle between each jar.
